The effect of quercetin on fertility of frozen-thawed ram epididymal spermatozoa

The aim of the present study was to evaluate the effects of quercetin as an antioxidant supplement on frozen-thawed ram epididymal sperm quality. Quercetin is a type of flavonoid antioxidant that is found in plants, with the ability to scavenge free radicals. Twenty testicles from mature rams were collected from a nearby slaughterhouse immediately after slaughter. Epididymal spermatozoa were recovered from the caudal of epididymides by injecting Bracket and Oliphant's (BO) medium retrogradely through the ductus deferens and extended with a tris egg-yolk-based extender and supplemented with 0, 5, 10, 15, 20, and 50 µg/mL quercetin. Following equilibration, the straws were frozen, and then plunged into liquid nitrogen. After thawing, optimized concentrations of quercetin were defined based on their viabilities and used to assess fertilization and developmental potential. The results showed that the viability of frozen-thawed spermatozoa significantly increased by using 5 and 10 µg/mL quercetin in the freezing extender. However, total and progressive motility of frozen-thawed spermatozoa were not affected by 5 and 10 µg/mL quercetin in comparison with control (0 µg/mL). The mean number of zygote, morula, and blastocyst stage embryos increased significantly by using 5 and 10 µg/mL quercetin compared with other frozen-thawed treatments(P <0.05). However, the blastocyst rate of fresh sperm was significantly higher (P <0.05). In conclusion, to improve the quality of frozen-thawed ram epididymal spermatozoa, 5 and 10 µg/mL quercetin appears to be an attractive option. Further studies are suggested to understand the synergistic effect of quercetin with other antioxidants to improve the ram freezing-thawing process.

Male reproductive parameters are often used for the functional examination and evaluation of predicted genetic values for future aspects. However, these traits are relatively reliable until the measurable effects are expressed on desired traits. Therefore, we aimed to associate the single nucleotide polymorphism (SNP) genotype of the investigated characteristics and reproductive loci. A total of 46 male dogs are divided into three age groups (I ≤ 3 years, n = 19; II 4-6 years; n = 19, and III ≥7 years, n = 8). The testis, scrotum and body weight, libido sexualis and ejaculation time for each fraction were monitored as functional traits, while the pH, fractional semen volume, motility, concentration, and abnormal and dead spermatozoa rate were recorded as spermatological traits. The Affymetrix Canine 127 K SNP genotyping array v2 (Affymetrix Inc., California, USA) was used for SNP genotyping. In the primary results, the scrotal circumference was found to be higher in group II compared to other groups (p < 0.05) and the lowest total abnormal spermatozoa rate was found in group I (p < 0.05). The normal spermatozoa rate was found to be significantly above the threshold in relation to the SNP in chromosome 17. In conclusion, this study represents an exciting first step towards SNP association with dog semen spermatological parameters. Future studies might be undertaken to evaluate this SNP region for gene-knockout and expression analysis and for fine mapping to validate and/or discover the exact position of the effect region.

In the presented study, the protective role of carnosic and gallic acid on post-thaw ram sperm acrosome and membrane integrity was examined, homocysteine (Hcy) and nesfatin levels were determined. In study semen samples from 6 Merino rams were used. Each ejaculate, split into five equal aliquots was diluted with extenders including 0.05 mM gallic acid (GA), 2 mM gallic acid, 0.05 mM carnosic acid (CA), 0.2 mM carnosic acid and no additive (control) at 37 °C cooled to 5 °C then frozen at nitrogen vapor. Freeze-thawed ram semen viability was achieved in gallic acid 2mM (57.13± 2.38%) group and statistical difference was found with control group (45.08±2.98%) (p<0.05). There was no statistical difference between the groups in HOS test analysis (p>0.05). The lowest level of homocysteine was obtained in the gallic acid groups (0.67±0.11 and 0.61±0.26 μmol/L) and was found statistically different with the control group (1.36±0.9 μmol/L) (p<0.05). No significant difference was found in nesfatin levels among groups (p>0.05). GA supplementation in ram semen extender has been determined to protect viability and HOS test parameters also to decrease homocysteine level.
