Relationship Between Platelet and Urinary 8‐Iso‐PGF2α Levels in Subjects With Different Degrees of NOX2 Regulation

Abstract: BackgroundUrinary 8‐iso‐PGF2α, a marker of oxidative stress, is influenced by the activation of NOX2. It is unclear if platelets 8‐iso‐PGF2α contribute to urinary 8‐iso‐PGF2α.Methods and ResultsIn a cross‐sectional study, platelet, urinary, and serum 8‐iso‐PGF2α were determined in subjects with downregulation (X‐linked chronic granulomatous disease [X‐CGD], n=25) and upregulation (type II diabetic patients [T2D], n=121) of NOX2 and 153 controls matched for sex and age. In diabetic patients (n=18), the above va… Show more

“…This finding was corroborated by analysis of in vivo platelet activation in obese subjects who, conversely, disclosed NOx2 upregulation coincidentally with a significant increase of both sCD40L and sP‐selectin. This is consistent with previous studies showing upregulation of NOx2 in patients with atherosclerotic risk factors such as those with hypercholesterolemia or diabetes …”

“…As previously shown by Carnevale et al [42], patients affected by diabetes mellitus, which is a model of high risk to development of cardiovascular complications, show higher levels of serum Nox2-dp and urinary, platelet and serum isoprostanes, with respect to healthy individuals and sNox2-dp (pg/ml) patients with hereditary deficiency of NOX2, as X-linked chronic granulomatous disease (X-CGD). So, as shown in the diabetic patients, in overall hypertensive patients, we have found a significant correlation between serum Nox2dp and urinary isoprostanes; moreover, in primary aldosteronism patients, we also found a significant correlation between serum Nox2-dp with plasma aldosterone levels and 24-h urinary excretion of aldosterone, and between plasma aldosterone and urinary isoprostanes.…”
