Relationship between Dietary Intake and Tissue Levels of Reduced and Total Vitamin C in the Nonscorbutic Guinea Pig

Abstract: The objective of this study was to determine the effect of a broad range of dietary intake levels of ascorbate on the distribution of both total and reduced ascorbate in guinea pig tissues. Young male Hartley guinea pigs were fed for 2 mo a modified Reid-Briggs purified diet containing five different levels of total ascorbate that provided 0.8-52 mg ascorbate/d. We also fed aged guinea pigs two different levels of ascorbic acid (1.5 or 60 mg/d) for 2 mo. Reduced and total ascorbate was measured in eye lens and… Show more

“…0.5-inch needle syringe. The samples were treated as previously reported [11]. An equal volume of 0.35 M perchloric acid with 0.1 mg/ml EDTA and 0.2 mM glutathione was added to the aqueous sample and then filtered through a 0.22-Ìm membrane by centrifugation (UFC30GV00, Millipore).…”

“…2.8. Determination of AA levels Ascorbic acid (AA) levels were estimated as previously described (Berger et al, 1989). In brief, homogenized (on ice in 0.35 M perchloric acid) samples were centrifuged for 3 min at 15,000 × g. A 400-μL aliquot of either the standard (AA) or sample was mixed with 100 μL of color reagent in a centrifugal analyzer tube.…”

“…The average daily as corbate supplementation was 0.9 mg. All unopened bags of diet were frozen at -2 0 0 C to ensure that ascor bate did not deteriorate as a result of storage. To ensure consistency and accuracy of the animals' daily ascorbate intake, each bag of guinea pig diet was ana lyzed for reduced and total vitamin C content utilizing methods described by Berger et al [ 12] and Roe and Keuther [ 17], All guinea pigs were provided free access to drinking water treated by reverse osmosis, double ion exchange, and ultraviolet irradiation.…”

“…The Low-C diet (~ 5 mg/day) provided di etary levels of ascorbate, which were adequate to prevent scurvy, while the High-C diet (74 mg/day) provided more than ten times the daily recommended requirement for guinea pigs [ 19], This is enough to saturate all tissues in the parental strain with ascorbate [11,12], Ascorbate levels were evaluated at the conclu sion of the experiment to determine the bio availability of ascorbate in organs of interest. Table 1 shows that the diets resulted in two groups of hairless animals with different as corbate status.…”
