Regulation of CXCL9/10/11 in Oral Keratinocytes and Fibroblasts

Abstract: Th1 and Th2 cytokines such as interferon-gamma (IFN-gamma ) , tumor necrosis factor- alpha (TNF-alpha ), and IL-4 are expressed in T-cell-mediated inflammation in the oral cavity. We tested the hypothesis that those cytokines may act on CXCR3-agonistic chemokines, T-cell recruiting factors, and on neighboring cells, including oral keratinocytes and fibroblasts. Human immortalized oral keratinocytes (RT7) and fibroblasts (GT1) after 24-hour stimulation with IFN-gamma showed increased mRNA levels of CXCL9 (600- … Show more

“…The T-cell–attracting chemokines CXCL9, 10, and 11 are known to be induced by IFNγ in various cell types, including ECs (Sauty et al , 1999; Kanda et al , 2007; Kanda and Watanabe, 2007; Ohta et al , 2008; Kawaguchi et al , 2009). Although CD40 stimulation salvaged the expression levels of CXCL9 , CXCL10 , and CXCL11 in HPV-positive ECs to similar levels found in noninfected ECs (Figure 4d–f), ELISA assays showed that hrHPV-positive ECs still secreted lower levels of CXCL9 and CXCL10 compared with noninfected ECs (Figure 4g and not shown).…”

“…CXCL9 is a T-cell chemoattractant (Gorbachev et al, 2007; Hertenstein et al, 2011), which is secreted by all kinds of cell types including neutrophils, monocytes, endothelial cells and fibroblasts (Taub et al, 1993). Studies have shown that in the setting of disease or stimulation with pre-inflammatory cytokines, the level of CXCL9 was increased (Coma et al, 2006; Ohta et al, 2008). C-X-C motif chemokine 13 (CXCL13), also known as B lymphocyte chemoattractant is a homeostatic chemokine that is constitutively expressed in secondary lymphoid tissue and involved in lymphoid organogenesis (Carlsen et al, 2004).…”

“…Primary cultures of human keratinocytes and fibroblasts from oral mucosal biopsy samples from healthy volunteers were prepared as previously described [8]. Informed consent for acquisition was obtained according to a protocol approved by the Ethical Committee of Hiroshima University.…”

“…We previously reported that oral keratinocytes and fibroblasts produce CXCL10, which is thought to attract immune cells to the site of oral inflammation [8]. The major function of CXCL10 in host defense against viral infection has been shown in CXCL10-/- mice, in which it impaired recruitment of effector T cells and NK cells to sites of infection/inflammation [9,10].…”
