“…Additional SSR markers were obtained through a search of GrainGenes (http://wheat.pw.usda.gov, verified 20 Jan. 2014) for markers that previously had been mapped to genomic regions of interest. The 12-μL reaction mixture consisted of 120 ng of template DNA, 1.2 μL 10´ Mg-free PCR buffer (New England Biolabs, Ipswich, MA), 1.0 unit of Taq DNA polymerase, 1.0 mM of MgCl 2 (Fermentas, Glen Burnie, MD), 200 µM dNTP (deoxyribonucleotide triphosphate, New England Biolabs, (PI 658064;Fowler, 1997), Norstar (CI 17735;Grant, 1980), Tiber (PI 517194; Kisha et al, 1992), Froid (CI 11957;DeNoma, 1990) and germplasm line Oregon Feed Wheat #5 (ORFW) (Skinner and Garland-Campbell, 2008b). The PCR primers were synthesized to include an M13-tail on the 5¢ end of the forward primer (Oetting et al, 1995).…”
