Regeneration of bulblets from twin scales ofCrinum macowanii in vitro

Abstract: Tissue culture techniques were applied to study the regeneration and growth of bulblets from bulb scale segments of Crinum macowanii Bak. (bush-or march lily) in vitro. Shoots were induced on twin scales taken from the basal plate region of flowering-size bulbs on Murashige and Skoog (MS)-medium containing 0 -2 0 m g l -~ NAA and BA and a modified MS medium (MMS medium) containing 1.25 mg !-~ ancymidoi (A-RestrU), 0.1 mg i-1 NAA and 0.1 mg 1-~ kinetin (ANK). Large bulblets could only be initiated on the latter… Show more

“…The use of growth retardants to induce corm formation has met with success in ornamentals such as Gladiolus (Ziv 1989(Ziv , 1990(Ziv , 1992Steinitz et al 1991), Crinum (Slabbert et al 1993), Crocus (Piqueras et al 1999) and Dierama (Madubanya et al 2006). In all species of Watsonia, a higher concentration of PAC (10 mg l -1 ) inhibited shoot and root growth and resulted in meristemoid formation, similar to that in Fig.…”

“…Addition of GA 3 significantly increased corm induction compared to the control, and storage organs were heavier, though this was not significant. This is intriguing since in all other Iridaceae that have been examined, anti-gibberellin agents promoted corm induction (Ziv 1989;Slabbert et al 1993;Piqueras et al 1999;Madubanya et al 2006). Although it is more frequent for GA 3 to inhibit storage organ formation (Le GuenLe Saos et al 2002), this promotion of corm induction by GA 3 is not unique to W. vanderspuyiae.…”

“…hybrids (Seabrook and Cumming, 1977), there was greater production of shoots in a growth medium without growth regulators. The same result was found for Crinum macowanii (Slabbert et al, 1993) and for the beginning cultivation phase of Eucomis comosa and Eucomis zambesiaca (Ault, 1995).…”

The same result was found for Crinum macowanii (Slabbert et al, 1993) and for the beginning cultivation phase of Eucomis comosa and Eucomis zambesiaca (Ault, 1995).Regarding the micropropagation of Amaryllidaceae, several studies emphasized the necessity of determining the optimal concentration of growth regulators required by each species in each phase of the in vitro culture. There are species that need higher concentrations of cytokinins (C谩zarez et al, 2010;Uranbey, 2010), whereas others require auxins (Pi帽a-Escutia et al, 2010) in equivalent concentrations (Azadi and KhoshKhui, 2007).…”
