Reduction in Desorption Mass Spectrometry: Multiple Protonation on Flavins without Charge Increment.

Abstract: Three flavin-containing compounds, namely, riboflavin, riboflavin 5ῐ-phosphate (flavin mononucleotide, FMN) and flavin-adenine dinucleotide (FAD, Scheme 1), were studied using matrix-assisted laser desorption ionization (MALDI), fast atom bombardment (FAB) and electrospray ionization (ESI) mass spectrometry. It was found that the isoalloxazine ring (flavin) was reduced in the positive-ion MALDI and FAB ionization processes producing unusual ions [Mῌ2] ῌῌ and [Mῌ3] ῌ in addition to the anticipated [Mῌ1] ῌ , w… Show more

“…12) As reported by Itoh et al, flavin-containing compounds, riboflavin, riboflavin 5῏-phosphate, and flavin-adenine dinucleotide, were reduced by MALDI and fast atom bombardment (FAB), whereas such reduction was not observed in the ESI mass spectrum. 13 ῌ ions were dependent on the matrix species used for MALDI. 13) Also, photo-induced dissociation followed by reduction was suggested in the MALDI-MS of nitrotyrosine, in which the NO 2 moiety is partially changed into an amine (NH 2 ).…”

“…16 It is postulated that protonation (by the matrix), with concomitant electron transfer in the gas phase and an energy contribution from UV-laser irradiation, could induce the hydrogenation of the double bond adjacent to nitrogen in the pyridine ring of PA, thus resulting in the production of …”

“…17 In the case of nitrotyrosine, the NO2 moiety is partially converted into NH2, probably due to reduction after photo-induced dissociation by UV laser irradiation. 18,19 Similarly, the S-nitrosylated cysteine, Cys(SNO), in peptides is completely converted to cysteine, Cys(SH), in MALDI.…”
