Reduced Pathology following Infection with TransgenicLeishmania majorExpressing Murine CD40 Ligand

Abstract: Leishmanization is the inoculation of live Leishmania into the host to vaccinate against subsequent infections. This approach has been largely discontinued due to safety concerns. We have previously shown that combining CD40 ligand (CD40L) with Leishmania antigen preferentially induces a type 1 immune response and provides some protection to vaccinated mice (G. Chen, P. A. Darrah, and D. M. Mosser, Infect. Immun. 69:3255-3263, 2001). In the present study, we developed transgenic L. major organisms which expres… Show more

“…The same thing was reported by Rolph and Kaufmann (2001), Chen et al (2001) and Field et al (2007) which uses a combination of CD40 and vaccine of heat-killed Listeria mononcytogenes and Leishmania major. The development of vaccines based on cell-mediated immunity was also reported by Lawrence et al (2012) in a vaccination using HKSA.…”

“…Another approach to stimulate CD40 signaling during immunization has been to utilize transgenic parasites that express CD154. Compared to wild-type parasites, infection with L. major promastigotes that express the extracellular domain of CD154 caused mild disease in susceptible BALB/c mice and afforded resistance against challenge infection to C57BL/6 mice [121]. …”

“…Susceptibility to L. major infection has been linked to a deficiency in the engagement between CD40 and CD40L. 8,[38][39][40][41][42][43][44] Furthermore, B7-mediated costimulation is required for the development of the early immune response following infection of either resistant or susceptible mice. 45 Therefore, activation and maturation of APCs with expression of MHC molecules, CD40 and CD80 on the surface of APCs is fundamental to the process of costimulation of T cells for an efficient response to L. major infection.…”
