Reduced Graphene Oxide-Based Impedimetric Immunosensor for Detection of Enterotoxin A in Milk Samples

Abstract: A simple, cheap, and less aggressive immobilization procedure for biomolecules using reduced graphene oxide (rGO) was employed to prepare an impedimetric immunosensor for detection of staphylococcal enterotoxin A (SEA) from Staphylococcus aureus in milk samples. The scanning electron microscopy, cyclic voltammetry, and electrochemical impedance spectroscopy (EIS) were used to monitor the single steps of the electrode assembly process. The glassy carbon (GC)/rGO platform detected the antigen-antibody binding pr… Show more

“…Moreover, ΔE p of 237 mV for the GC/rGO-EDC-NHS/Ab electrode and 358 mV after the SARS-CoV-2 antigen is incubated are observed. This increase in the ΔE p was also observed in the antigen-antibody binding procedures of different types of electrochemical immunosensors [ 19 , 22 ]. In addition, Figure 2 C showed that the decrease of currents generated by the [Fe(CN) 6 ] 4− / 3− system observed in the cyclic voltammograms after SARS-CoV-2 antigen binding at the GC/rGO-EDC-NHS/Ab/BSA electrode has a clear correlation with the concentration of the antigen.…”

“…Our research group used the reduced graphene oxide (rGO) to immobilize enzymes for the preparation of enzymatic biosensors to monitor glucose during the second-generation ethanol production [ 16 ], to analyze neurotransmitters in urine and plasmatic serum samples [ 17 ] and to determine pesticides in food [ 18 ]. rGO was also employed as a platform in the development of an electrochemical biosensor for detection of staphylococcal enterotoxin A in milk samples [ 19 ].…”

“…The research shows that antibodies and antigens have poor electrical conductivity, which could prevent the external redox mediator ([Fe(CN) 6 ] 3−/4− ) electrons’ transfer toward the electrode surface [ 20 ]. The EIS has been demonstrated to analyze the conductivity of the modified electrode.…”

“…Hence, the appropriate design and optimization of IDME is crucial for the detection performance. In addition, Au has been widely applied for IDME construction due to the simple fabrication and ease in forming self-assembled monolayers [ 20 ]. To pursue a higher sensitivity of a bioimpedance sensor, it is necessary to increase the amount of antibody as much as possible on the surface of an electrode.…”

“…In addition, exploring methods for performance improving, application diversification and high reliability is also an adjoin research proposition, which lead most aspects of techniques to upgrade and even fuse. For these goals, many kinds of tentative studies have been done, such as the metal-pasted thread as the important conductive medium, the graphene-based conductive medium [12,13], and special synthetic textile materials as substrates. Certainly, the use of novel materials will push the fabrication techniques to make changes.…”
