Reactive Oxygen Species, Cell Growth, and Taxol Production of Taxus cuspidata Cells Immobilized on Polyurethane Foam

Abstract: Dynamic changes in reactive oxygen species (ROS) of Taxus cuspidata cells immobilized on polyurethane foam were investigated and the relation between ROS content and taxol production was discussed. Immobilization shortened the lag period of cell growth and moderately increased H2O2 and O2-* contents inside the microenvironment within the first 15 d. After 20 d, excessive production of H2O2 and O2-* was observed accompanied by marked increases in membrane lipid peroxidation and cell membrane permeability. The t… Show more

“…Meanwhile the microenvironments were influenced by multiple factors, such as restriction by reticulated foam, nutriment transportation and oxygen diffusion (Yin et al, 2005), so the endogenous NO was produced differently in different layers in a long term. Fig.…”

“…The fungal elicitor and ultrasound (Wu and Lin, 2003) improved the Taxol production accompanied with increase of PAL activity (Yuan et al, 2001;Dixon and Paiva, 1995). As PAL was also the key enzyme in forming the side chain during Taxol biosynthesis, the increased PAL activity promoted Taxol production in immobilized T. cuspidata cells (Yin et al, 2005). Walker first illustrated that a phenylalanine aminomutase with high similarity to PAL was involved in taxane side chain synthesis (Walker and Floss, 1998).…”

“…This specific combination was selected as it has been shown to be effective in the recovery of taxanes from yeast cell factories (Santoyo-Garcia et al 2023 ). As demonstrated in previous studies, the total concentration of the resins in the culturable cells is a critical variable for cell growth (Yin et al 2005 ; Santoyo-Garcia et al 2022 ). For that reason, treatment A configuration (0.5, 1 and 1.5% w/v of HP-20, XAD7 and HP-2MG, respectively) of resins at different total concentrations (1.5, 3 and 6% w / v ) was utilised to test cell growth and taxanes production in VSCs.…”

“…The major oxygen species responsible for these oxidative stresses are hydrogen peroxide (H 2 O 2 ), the free radical superoxide anion (O 2- ) and the hydroxyl radical (OH - ) [6]. Among the ROS, H 2 O 2 has been implicated as a cellular toxin [19,61]. Since H 2 O 2 has remarkable membrane permeability [23], intracellular H 2 O 2 can induce detrimental effects in cells [20,41,42,49].…”
