The aim of this study is to assess the Xpert MTB/RIF assay for diagnosis of the Mycobacterium tuberculosis complex in clinical samples and to compare the results by reference to the diagnostic method, Bactec MGIT 960. Materials and methods:A total of 7407 samples were included from patients not primarily suggesting pulmonary or extrapulmonary tuberculosis (TB), collected from patients required to be screened for TB and excluding TB diagnoses since it was a differential diagnosis. Also included were a total of 411 samples from patients primarily suggesting pulmonary or extrapulmonary TB.Results: In the first group, 152 of 7407 samples yielded positive results with the Bactec MGIT 960, 131 (1.77%) were found positive with Löwenstein-Jensen medium, and 295 (3.99%) were found positive with Ziehl-Neelsen staining. In the second group, 24 (5.8%), 17 (4.1%), and 28 (6.8%) of 411 samples were found positive. Xpert MTB/RIF [27 (6.6%) of 411 samples] detected 3 additional samples as positive, and these 3 cases were clinically compatible with TB. Conclusion:The Xpert MTB/RIF assay shows superior performance for the diagnosis of TB. Its usefulness in culture-negative patients and the best method for integrating this diagnostic method into current tuberculosis diagnostic algorithms both need further study.

The aim of this study was to investigate the effects of various extracts obtained from Pistacia eurycarpa Yalt. on the exact time of appearance of the colonies of Mycobacterium tuberculosis on solid media. For this purpose, the fatty acid compositions of the fixed oils were analyzed using Gas ChromatographyMass Spectrometry (GC-MS) in the fruit and peels. The fixed oils and the water-soluble extracts obtained from the plant as well as the fatty acids were added either separately or together on the surfaces of the Löwenstein Jensen and Middlebrook 7H11 agar media. Their effects on growth time of 40 strains of M. tuberculosis complex were investigated. Therefore, all of water-soluble extracts accelerated the growth of the mycobacteria in a statistically significant manner in both media. It was observed that the fixed oils and the fatty acids individually did not cause any significant effect on growth duration of the M. tuberculosis strains. Finally, the water-soluble extracts of P. eurycarpa fruit and its peel significantly accelerated the growth of M. tuberculosis by shortening the growth duration of M. tuberculosis at least by one third.

ABSTRACT:A Mycobacterium DNA extraction protocol utilizing SPION developed and compared with the conventional and commercial kit method. The quality, quantity, and feasibility of the recovered DNA from all four different methods towards various molecular techniques were observed. In our work, morphology, bonding and composite of SPION is determined by various characterization techniques such as XRD, FT-IR, VSM, UV-vis spectroscopy and FE-SEM. XRD analysis confirms the crystallite size and phase purity of the synthesized material. The average crystallite size obtained were ~ 9.7 nm for SPION, prepared by a Co-precipitation method. FT-IR spectrum also confirms the formation of SPIONs. UV-vis spectrophotometry and gel electrophoresis analysis resulted in highest MTB DNA yield (985.3 ng/µl) and the purity was measured by the ratio A260 / A280 = 1.89. The comparative study suggested that the protocol developed in this report is a unique cost effective and highly pure. Further, Real-time PCR also confirms the superiority of our technique.
