RAPD and isoenzyme analysis of genetic variability in three allied species of catfish (Siluriformes: Pimelodidae) from the Tibagi River, Brazil

Abstract: Genetic variability in three species of Pimelodidae cat®sh (Iheringichthys labrosus, Pimelodus maculatus and Pimelodus cf. absconditus), collected at four sites in the Tibagi river-basin: Sertaneja, Londrina (lower Tibagi), Sapopema (middle Tibagi) and Tibagi (upper Tibagi) was analysed using RAPD and isoenzymatic techniques. Iheringichthys labrosus had the greatest genetic variability showing a proportion of mean polymorphic loci (P Å m) of 53.7%. The dendrogram showed that the individuals from the locations … Show more

“…annectans (n=203) and Eg. pleosticta (n=269) was performed as in Freiria et al (2012), while DNA from males of Eulaema cingulata (n=319), Euglossa iopoecila (n=271) and Euglossa truncata (n=42) was obtained according to Almeida et al (2001), with minor modifications, described as follows: parts of the body (legs or thorax) were placed in micro-centrifuge tubes containing 300 μL extraction buffer (500 mm Tris-HCl, 20 mm EDTA, 10 mm NaCl, pH 8.0 and 1 % SDS) and 5 μL K proteinase (20 μg/μL). Using small scissors, the body parts of bees were cut, held in a water bath at 63°C for two hours and then subjected to phenolchloroform washes.…”

“…Random amplified polymorphic DNA (RAPD) analysis, a simple and straightforward PCR-based technique (Williams et al, 1990), employs a class of markers increasingly used to evaluate genetic variability and structure in a variety of organisms (Kimberling et al, 1996;Almeida et al, 2001;Allnutt et al, 2003).…”

“…DNA was isolated from a few milligrams of lyophilized tissue using a protocol developed by Shiozawa et al (1992), with some modifications (Almeida et al 2001). DNA quality and concentration were evaluated via electrophoresis in agarose gels following staining with SYBR Safe (Invitrogen, CA, USA).…”
