A fully-automated on-line oxidation column-switching HPLC system has been developed for the determination of endogenous melatonin in human plasma and saliva. This HPLC system consists of four processes. In the first step, melatonin is fractionated using a reversed-phase C4 column (Proteonavi, 75 mm × 1.0 mm i.d.). In the second step, the obtained melatonin fraction is on-line collected, and oxidized to a highly-fluorescent compound, N- [(6-methoxy-4-oxo-1,4-dihydroquinolin-3-yl)methyl]acetamide (6-MOQMA), by mixing with hydrogen peroxide under alkaline conditions. In the third step, the produced 6-MOQMA is concentrated, and the oxidation reagents are removed using an alkaline resistive reversed-phase column, Asahipak ODP (35 mm × 1.0 mm i.d.). In the final fourth step, the 6-MOQMA is determined by a microbore-ODS column packed with ultrafine particles (CAPCELL PAK C18 IF, 250 mm × 1.0 mm i.d.). The limit of detection of melatonin using this system is about 200 amol/injection, and the determination of endogenous melatonin in a small volume of human physiological fluids, such as 100 μL of plasma and 300 μL of saliva, was successfully accomplished.

This chapter attempts to step backward in time and space, to consider evolution in the context of chronobiology and chronomics and to then focus on how this science, pioneered by Professor Halberg and his team at the University of Minnesota, can bring improved health benefits to mankind. Although being mindful of this old yet formally modern science, care is taken to include many practical considerations that have a tendency to be sometimes undervalued in research.Evolution continues under the omnipresence of gravity, changing electromagnetic forces, time, space, and Earth's climate and environment as an ecological biogeographical phenomenon that took place extremely slowly in geological time but is now undergoing change at an extraordinary pace in recent times of human evolution and adaption to their environment [1]. Already about 1 out of 3 billion people go hungry [2] (and currently in overall decline) and by 2050 the estimated population is about 9 billion [3]. Great challenges to reduce poverty and hunger lie ahead: people need to be fed but safely, the risk of inflammatory diseases such as gastrointestinal and cardiovascular disorders needs to be reduced: and one potential area of research concerns chrononutrition. It is by understanding the chronobiological science that underpins health that will enable such goals to be reached.

Quantitative determination of melatonin in milk by LC-MS/MS

A rapid, reliable and sensitive LC-MS/MS method for the determination of melatonin in milk was developed and validated. Sample was extracted with dichloromethane and cleaned by passing through Chem Elut solid phase extraction cartridge. The solvent was evaporated to dryness, reconstituted with methanol and analysed by LC-MS/MS on Agilent zorbax Eclipse XDB C-18 rapid resolution analytical column. The analytical procedure was found to be accurate, precise and linear. The method accuracy was 92.2 % (range 90.06-94.58) and the mean precision was 1.55 % and the calibration was linear for 1 to 150 pg mL −1 (R 2 >0.99), the lowest limit of quantification (LLOQ) was 1 pg mL −1. 7-D Melatonin (7-DM) was used as an internal standard. This method was proved to be a promising method for the determination of melatonin for market milk and human milk samples.
