Pyridone Luciferins and Mutant Luciferases for Bioluminescence Imaging

Abstract: New applications for bioluminescence imaging require an expanded set of luciferase enzymes and luciferin substrates. Here, we report two novel luciferins for use in vitro and in cells. These molecules comprise regioisomeric pyridone cores that can be accessed from a common synthetic route. The analogues exhibited unique emission spectra with firefly luciferase, although photon intensities remained weak. Enhanced light outputs were achieved by using mutant luciferase enzymes. One of the luciferin-luciferase pai… Show more

“…Replacement of the sulfur atom in the thiazoline ring of d -luciferin and AL with carbon or selenium resulted in luminescence wavelengths of 547 nm and 600 nm, respectively (Compound 47 , 48 ) [ 43 , 44 ]. Substitution of pyridone ring instead of the benzothiazole ring showed a luminescence wavelength of 570 nm or 530 nm (Compound 49 , 50 ) [ 45 ].…”

“…Furthermore, luciferase signals can be easily quantified [ 64 ]. Being widely used for in vitro analysis, the intracellular functions of these bioluminescent systems can be hindered by their overall dim signals, limited choice of wavelengths, and due to the limited availability of the luciferin substrates [ 65 ].…”

“…[2628] Both blue- and red-shifted analogs have been synthesized in recent years, although most remain weak emitters with native luciferases. [26,27,29] Engineering enzymes to better process the modified analogs—and thus recover light intensity—has been successful in some cases. [27,29,30]…”
