Purification and optimization of pink pigment produced by newly isolated bacterial strain Enterobacter sp. PWN1

Abstract: Pigment-producing bacteria were isolated from kitchen wastewaters of the National Institute of Technology, Rourkela. A pink non-virulent bacterial strain PWN1 was selected based on the India Ink Broth and Coomassie Brilliant Blue (R-250) dye assay. According to morphological and biochemical characterization, the strain PWN1was a Gram-negative, rod-shaped, motile, non-coliform bacterium and could utilize only glucose and adonitol as sole carbon source. The pigment was found to be a growth-associated product, an… Show more

“…The specific pigment production and the specific growth rate were calculated according to the following equations (Equations (2) and (3)): where μ is the specific growth rate, X is the cell biomass concentration (g/L), and t is the specific time (h). where P is the concentration of the extracted pigment (g/L) at time t (d) and X is the cell biomass concentration (g/L) [ 2 ].…”

“…Bacteria were considered as the ideal pigment-producing source because of several advantages, namely the short and rapid life cycle, easy maintenance or preservation of bacterial cultures, scalability for industrial use, the ability to adapt to environmental changes, the ability to grow on cheap, easily available media, simple extraction techniques, and renewable sources that can reduce the pigment production cost [ 2 ].…”

“…The process was repeated until complete pigment extraction was achieved. The pigment extract was evaporated at 40 °C by a rotary evaporator to dryness 82 . The extract was dissolved in petroleum ether and mixed with methanol in a separating funnel, and vigorously shaken for 10–15 min to allow the two liquids to separate.…”

“…Candida species transform from a commensal to a pathogenic form as a result of local, systemic, and/or host-related predisposing factors, which favor the progression of the disease. The systemic elements affect the host's defense mechanisms while the local factors provide a suitable oral environment for the production of biofilms (4,5).…”
