Pullulanase from Aerobacter aerogenes is a glucosidase specifically splitting a-1,6-glycosidic bonds. A polymeric substrate for the pullulanase is the linear a-glucan pullulan from Pullularia This a-glucan consists of maltotriose units linked together by a-l,6-glycosidic bonds. This means that the polysaccharide is degraded by pullulanase to yield exclusively maltotriose. There are two possibilities of enzyme action : a) either the hydrolysis starts from one end of the linear chain directly setting free maltotriose ; b) or random splitting occurs to give larger units of varying molecular weight which will Snally be degraded to maltotriose.The decision between these two alternatives could be made by gas liquid chromatography. The quantity of liberated maltotriose rises linearly with the reaction time, which proves that hydrolysis occurs from the end of the substrate chain. This method using gas liquid chromatography as an analytical tool may also be applied to monitor the action of other hydrolytic enzymes, as it could be shown for a-and p-amylase. pulluhrm.Die Pullulanase ist eine Glucosidase, die a-1,6-glykosidische Bindungen in Oligo-und Polysacchariden spaltet, wenn bestimmte weitere strukturelle Voraussetzungen gegeben sind [l-31. Das Enzym wurde in einem Stamm von Aerobacter aerogenes entdeckt [l], der auf Pullulan als einziger Kohlenstoffqueue wachst, aus Kulturen dieses Stammes isoliert, bis zur Einheitlichkeit gereinigt und kristallisiert [4,5]. Zur weiteren Charakterisierung des Enzyms untersuchten wir verschiedene Aspekte der Wechselwirkung mit dem Substrat mit dem Ziel, dieses Protein nicht nur auf Grund der Spezifitat, sondern auch der Wirkungsweise in die Gruppe der bekannten a-Glucanasen einzuordnen. Als Substrat diente fiir diese Versuche Pdulan, ein lineares Glucosepolymeres, das aus ca. 480 a-1,6-verkniipften Maltotrioseeinheiten aufgebaut ist [6,7]
