The effects of cyclodextrins and derivatives on the activity and structure of pullulanase were investigated in this study. Our results showed that cyclodextrins and derivatives decreased the activity of pullulanase. This decrease was attributed to the interaction between the hydrophobic cavities of cyclodextrins and pullulanase. The hydrophobic cavity was confirmed to encapsulate the groups of pullulanase molecules by the addition of competitive guests. The results obtained from fluorescence spectroscopy analysis showed that β-CD showed more efficient interactions with pullulanase molecules and the side chain groups of cyclodextrin significantly prevented the interaction between the hydrophobic cavities of β-CD and pullulanase molecules. These findings suggest that the geometric dimension of hydrophobic cavities was crucial for matching between cyclodextrins and pullulanase and steric hindrance caused by side chains led to the decrease of the interaction.

a-, p-und y-Cyclodextrin sind aus sechs, sieben bzw. acht Glucoseeinheiten bestehende, ringformige Oligosaccharide, die in groBtechnischem MaBstab aus Starke gewonnen werden konnen. Sie bilden EinschluBverbindungen mit kleineren Molekiilen, die in ihren 5-8 A weiten Hohlraum hineinpassen. Diese (kristallinen) Komplexe sind fur die Forschung interessant, da sie im Gegensatz zu den klassischen Clathraten bereits in waBriger Losung existieren und sich an ihnen die in biologischen Systemen so wichtigen hydrophoben Wechselwirkungen studieren lassen. Cyclodextrine dienen auBerdem als Modelle fur die polymere Starke und, als kristalline Polyiodid-Komplexe, fur die ,,blaue Iod-Starke". Da Cyclodextrine mehrere chemische Reaktionen katalysieren, sind sie und ihre Derivate mit funktionellen Gruppen wertvolle Enzymmodelle. Bei der Herstellung von Pharmaka, Schadlingsbekampfungsmitteln, Nahrungsmitteln und Toilettenartikeln lassen sich Cyclodextrine vorteilhaft verwenden, da die in ihnen eingeschlossenen (mikro-verkapselten) Wirk-und Aromastoffe vor Licht und atmospharischen Einflussen geschutzt sind und sich in pulverisierter Form leicht handhaben und aufbewahren lassen. In Wasser schlecht losliche Substanzen werden in Gegenwart von Cyclodextrinen besser loslich, Cremes und Emulsionen konnen stabilisiert, das Wachstum und der Ernteertrag von Getreide gesteigert werden. Cyclodextrine lassen sich fur vielfaltige Zwecke chemisch modifizieren; polymerisiertes oder an polymere Trager gebundenes Cyclodextrin wird bereits in der Gelinklusions-und Affinitatschromatographie verwendet.

a-, p-, and y-Cyclodextrins are cyclic oligosaccharides consisting of six, seven, or eight glucose units, which can be obtained on a large scale from starch. They form inclusion compounds with smaller molecules which fit into their 5-8 A cavity. These (crystalline) complexes are of interest for scientific research as, contrary to the classical clathrates, they exist in aqueous solution and can be used to study the hydrophobic interactions which are so important in biological systems. Cyclodextrins also serve as models both for polymeric starch and, in the form of their polyiodide complexes, for "blue iodine-starch". As cyclodextrins catalyze several chemical reactions they and their functionalized derivatives provide useful enzyme models. Cyclodextrins can be used to advantage in the production of pharmaceuticals, pesticides, foodstuffs, and toilet articles-the (micro-encapsulated) active and aromatic substances enclosed within them are protected from the effects of light and atmosphere and can be easily handled and stored in powder form. Substances which are not very soluble in water become more soluble in the presence of cyclodextrins-creams and emulsions can be stabilized, and the growth and yield of grain harvests can be increased. Cyclodextrins can be chemically modified for many different purposes; polymerized cyclodextrin or cyclodextrin bound to a polymer carrier have already been employed in gel inclusion and affinity chromatography.
