Viele natiirlich vorkommende und synthetische Liganden konnen Nucleinsauren selektiv erkennen, Oligonucleotide aber zeigen die hochste Erkennungsspezifitat. Sie binden unter Bildung von Watson-Crick-Wasserstoffirucken an eine komplementare einstrangige Sequenz. Des weiteren konnen sie in der groBen Furche von Doppelhelix-DNA bestimmte Sequenzen erkennen, indem sie mit Purinbasen der Watson-Crick-Basenpaare Hoogsteen-oder reverse Hoogsteen-Wasserstoffirucken bilden, wobei eine Tripelhelix cntsteht. An der sequenzspezifischen Wechselwirkung sind hauptsachlich Homopurin . Homopyrimidin-Sequenzen in der Doppelhelix beteiligt. Es ist noch immer eine Herausforderung fur Chemiker, den Bereich der Erkennungssequenzen zu erweitern. Es sind sowohl thermodynamische als auch kinetische Parameter fur die Bildung von Tripelhelices bestimmt worden. Diese Parameter zeigen beispielsweise, daB die Bildung einer Tripelhelix vie1 langsamer ist als die einer Doppelhelix. Nuclease-resistente Oligonucleotide, die aus dem a-Anomer von Nucleosiden (anstatt aus dem natiirlich vorkommenden fl-Anomer) dargestellt werden, bilden mit doppelstringiger DNA ebenfalls Tripelhelices. Tripelhelix-bildende Oligonucleotide konnen mit DNA-intercallerenden Agentien verkniipft werden, wodurch sie eine hohere Bindungsaffinitat erhalten. Oligonucleotide konnen auch durch Reagentien modifiziert werden, die durch chemische oder photochemische Aktivierung in der Zielsequenz irreversible Reaktionen induzieren. Es konnen so kunstliche Nucleasen entwickelt werden, die eine hohe Sequenzspezifitat selbst fur Megabasen-DNA aufweisen. Tripelhelix-bildende Oligonucleotide konnen ferner zur selektiven Steuerung der Genexpression verwendet werden. Binden sie an regulatorische Bereiche bestimmter Gene, so konnen sie die Aktivierung (oder Repression) der Transkription verhindern. Werden sie in der Nahe oder downstream von der Stelle gebunden, an der die Transkription beginnt, so kann die Fortsetzung des Ablesevorgangs gehemmt werden. Es ist vorstellbar, daI3 sich neue genblockierende Agentien entwickeln lassen, die zur therapeutischen Behandlung genetischer Storungen eingesetzt werden konnen.

Nucleic acids can be selectively recognized by a large number of natural and synthetic ligands. Oligonucleotides provide the highest specificity of recognition. They can bind to a complementary single-stranded sequence by forming Watson-Crick hydrogen bonds. They can also recognize the major groove of double-helical DNA at specific sequences by forming Hoogsteen or reverse Hoogsteen hydrogen bonds with purine bases of the Watson-Crick base pairs, resulting in a triple helix. Triple-helix formation through oligonucleotide binding to DNA is a sequencespecific interaction involving primarily homopurine . homopyrimidine sequences in the double-helical target. Extending the range of recognition sequences remains a challenge to chemists. Both thermodynamic and kinetic parameters for triplex formation have been determined. These parameters indicate, for example, that triple-helix formation is a much slower process than duplex formation. Nuclease-resistant oligonucleotides synthesized with the aanomers of nucleosides (instead of the natural p-anomers) also form triple helices with doublestranded DNA. Triple-helix-forming oligonucleotides can be modified, for example, by attaching DNA intercalating agents to enhance their binding affinity. They may also be modified with reagents that induce irreversible reactions in their target sequence upon chemical or photochemical activation. Thus, artificial nucleases can be developed with very high sequence specificity on megabase-size DNA. Furthermore, triple-helix-forming oligonucleotides can be used to selectively control gene expression. When bound to the regulatory region(s) of specific genes they may prevent activation (or repression) of transcription. When binding occurs near or downstream from the transcription initiation site, elongation of the transcript may be inhibited. Therefore, the potential exists for developing new gene-blocking agents with therapeutic applications in the treatment of gene disorders.

Electrochemistry of nucleic acids (NAs) is a booming field offering sensors for DNA hybridization and DNA damage. Since the middle of the 1990s, when the development of the DNA sensors began, the number of papers in this field is steadily increasing. Electroactivity of DNA and RNA was reported for the first time in 1958. Using oscillographic polarography at controlled AC (OP, AC chronopotentiometry) and a dropping mercury electrode it was shown within several years that OP signals respond to changes in DNA structure and can be used to study DNA denaturation, renaturation, premelting and DNA damage. During the first three decades a number of important findings were done, including introductions of covalently bound electroactive labels in DNA, DNA-modified electrodes, application of solid electrodes, etc., which are now used in the development of the NA sensors. On the other hand a lot of knowledge obtained in the first three decades has remained largely unknown to the present generation of researchers.
