Protective effects of β-glucan extracted from Agaricus brasiliensis against chemically induced DNA damage in human lymphocytes

Abstract: beta-Glucans (BGs) are polysaccharides that are found in the cell walls of organisms such as bacteria, fungi, and some cereals. The objective of the present study was to investigate the genotoxic and antigenotoxic effects of BG extracted from the mushroom Agaricus brasiliensis (=Agaricus blazei Murrill ss. Heinemann). The mutagenic activity of BG was tested in single-cell gel electrophoresis assays with human peripheral lymphocytes. In addition, the protective effects against the cooked food mutagen 3-amino-1-… Show more

“…In this study, we show that ABM extract possesses direct chemotactic activity for neutrophils. In effective components, ABM extract is also known to contain β-glucan, proteoglycan, and ergosterol [18], which are potent stimulators of macrophages, PMNs and NK cells [19][20][21].…”

“…A. brasiliensis-derived PLS obtained at LP/DQOI/ UFC (Gonzaga et al, 2005) was dissolved in Dulbecco΄s Modified Eagle Medium (DMEM), filter-sterilized and kept at 4ºC for a maximum of 72 hrs. The β-glucan was isolated from the polysaccharide-peptide assemblage extracted from A. brasiliensis (Angeli et al, 2006) and supplied by Laboratório de Polímeros, Departamento de Química Orgânica e Inorgânica, UFC, Brazil. The compounds were dissolved in DMEM, submitted to sterilization by ultra-filtration in 0.2 μm pore size membrane, and stored at -20°C.…”

“…Worldwide, they have attracted attention as a functional food and a source for the development of new immunomodulator compounds due to their dietary and medicinal benefits (Ooi and Liu 2000;Bertozzi and Kiessling 2001;Wasser 2002). The polysaccharide is localized in the intermediate layer of the cell wall, adjacent to the plasma membrane, with the function of maintaining the rigidity and shape of the cell (Angeli et al 2006). Carbohydrate is a major component in mushrooms, and its total content ranges from 35 % to 70 % dry weight, with variations in different species (Manzi et al 1999;Cheung 2010).…”
