Protection Duration of COVID-19 Vaccines: Waning Effectiveness and Future Perspective

Abstract: The coronavirus disease 2019 (COVID-19) vaccines have very successfully decreased the disease risk as we know; some key information remains unknown due to the short development history and the lack of long-term follow-up studies in vaccinated populations. One of the unanswered issues is the protection duration conferred after COVID-19 vaccination, which appears to play a pivotal role in the future impact of pathogens and is critical to inform the public health response and policy decisions. Here, we review cur… Show more

“…The seroconversion rate was higher with natural infection but the antibody titer was lower than that occurred after vaccination. The lower antibody titer may be due to the waning of the antibody over time ( 3 , 37 ). It is speculated that whole virus-based vaccines may induce the robust seroconversion and elicitation of antibody titer as reflected in our finding ( Table 2 ) ( 10 ).…”

“…Although there are data regarding the duration and persistence of antibody and T cell responses following many COVID‐19 vaccines such as BNT162b2 (Pfizer–BioNTech), AZD1222 and mRNA‐1273 14 , 15 there are limited data regarding the persistence of immune responses following Sinopharm/BBIBP‐CorV. 16 It is crucial that severe disease and hospitalizations due to COVID‐19 are curtailed to a level that it no longer becomes a global threat.…”

“…11 Based on waning of neutralizing antibody responses and efficacy for certain COVID-19 vaccines with time, booster doses are currently been offered to older individuals and individuals in high risk categories in different countries. 12,13 Although there are data regarding the duration and persistence of antibody and T cell responses following many COVID-19 vaccines such as BNT162b2 (Pfizer-BioNTech), AZD1222 and mRNA-1273 14,15 there are limited data regarding the persistence of immune responses following Sinopharm/BBIBP-CorV. 16 It is crucial that severe disease and hospitalizations due to COVID-19 are curtailed to a level that it no longer becomes a global threat.…”

“…Further, studies suggest that the defensive antibodies were reduced to approximately onehalf of the initial state after five months from the date of vaccination [17][18][19]. It causes more people to be vulnerable to infection.…”
