Properties of Selected Hemicellulases of a Multi-Enzymatic System fromPenicillium funiculosum

Abstract: A multi-enzymatic system from Penicillium funiculosum displayed -L-arabinofuranosidase, endo-1,4--Dxylanase, -D-xylosidase and endo-1,3-1,4--D-glucanase activities at high levels over a wide acidic pH range of 2.0 to 5.5. Moreover, the pH stability was particularly extended over the wide range of pH of 2.0 to 8.0 with endo-1,3-1,4--D-glucanase and endo-1,4--D-xylanase; however, -L-arabinofuranosidase and -D-xylosidase exhibited higher stability in the pH range of 2.0 to 5.5. The results indicate that the optim… Show more

“…One FTU is defined as the amount of enzyme that liberates one micromole of inorganic ortho-phosphate from phytic acid per minute at pH 5.5 and 37°C (Engelen et al 1994). In addition to endo-β-1,4-xylanase, endo-β-1,3(4)-glucanase and phytase activities, other enzymes (cellulases, pectinases, mannanase, and others) are also present and active in the preparation (Karboune et al 2008(Karboune et al , 2009. The presence of the enzyme preparation in the supplemented diets was confirmed by analyzing the corresponding xylanase and phytase activities.…”

“…the amount of enzyme, which hydrolyzes wheat arabinoxylans, reducing the viscosity of the solution to give a change in relative ñuidity of 1 arbitrary unit-min"û nder the assay conditions (pH 5.5 and 30°C) for xylanase activity, and the quantity of enzyme, which hydrolyzes barley ß-glucan (bound to a chromophore), releasing ethanol-soluble oligomers to give an absorbance of 0.820 units at 590 nm for endo-ß-l,3(4)-glucanase activity. In addition to endo-ß-l,4-xylanase and endoß-l,3(4)-glucanase activities, other enzymes (cellulases, pectinases, mannanase, and others) are also active in the preparation (Karboune et al, 2008(Karboune et al, , 2009. “…Penicillium [20] has been proved to be an excellent producer of cellulases and hemicellulases for industrial application as well as the genera Aspergillus and Trichoderma. Several endo-b-1,4-glucanases from Penicillium pinophilum KMJ601 [19], P. purpurogenum [22], and P. echinulatum [32] have been reported, but all of them only have the ability to hydrolyze the b-(1 ?…”
