Production of α-Amylase and Amyloglucosidase by the Fungus Cylindrocladium sp. in Semi-solid State Fermentation

Abstract: A mylases are very important and have a wide range of applications. Semi-solid state fermentation is increasingly used for the production of enzymes, since it requires simple technology and costs are low. In this regard, it is worth emphasizing the importance of rice as a substrate, due to its readily availability and low cost. This study evaluated the ability of the fungus Cylindrocladium sp. to produce α-amylase and amy loglucosidase. The studies were carried out with the fungus strain Cylindrocladium sp. wi… Show more

“…The inhibition activity of fucoidan against amyloglucosidase was determined by measuring the reducing power of released reducing sugar from soluble starch according to the method by Onofre et al (2012) with slight modification. The working enzyme solution was prepared freshly prior to use by diluting the stock amyloglucosidase solution (Megazyme, Ireland) to 6.5 U/ml in sodium phosphate buffer (SPB, 0.1 M, pH 6.9).…”

“…Reducing sugars were determined by reaction with 3,5-dinitrosalicylic acid (DNS) [18]. In a basic medium and at high temperature, this acid becomes 3-amino-5-nitrosalicylic acid, developing a red color and absorbing light at 540 nm.…”
