Production of mycophenolic acid by fungi of the genus Penicillium link

Abstract: Out of 36 strains of fungi of the genus Penicillium , some of which were isolated from ancient permafrost, 14 strains synthesized mycophenolic acid (MPA). Maximal (over 500 mg/l) accumulation of MPA in culture liquid was observed in P. brevicompactum strains (VKM F-457, VKM F-477, and VKM F-1150). This was the first study to detect MPA in representatives of the species P. rugulosum ; in three strains of this species , the level of MPA accumulation exceeded 300 mg/l. The time course of the synthesis of MPA by P… Show more

“…Results are presented in Table 2. Naphtalene derivatives are common as constituents of fungi (Frisvad et al 1998;Vinokurova et al 2005) andechinoderms (Bandaranayake, 2006). Conventional synthesis of naphtalenoids is a difficult task; therefore, isolation of compound 1 in the present study, with 45.34% yield from the extract (0.3175 mg.L -1 ) is very noteworthy making this compound a suitable starting compound for biotechnological use as a new drug lead, since its large scale production is feasible.…”

“…However, further structure elucidation using various NMR experiments confirmed the structure of 6 instead. This was surprising as compound 13, a proposed key intermediate in the biosynthesis of downstream metabolites such as 7, 8, and 10, was previously tentatively identified in different Penicillium cultures (8,10). By using a comparable instrumental setup with a similar chromatographic separation method, host and deletion strains of DS54555 were screened for production of 13.…”

“…Additionally, C-2 (␦ C ϭ 146) in the 13 C HMBC spectrum indicated a double bond between N-1 and C-2, which was supported by the chemical shift of N-1 (␦ N ϭ 280) in the 15 N HMBC spectrum. As compound 13 was reported from various Penicillium species such as Penicillium albocoremium (8), Penicillium glandicola (9), and P. chrysogenum (10) and proposed as a precursor of 7, host and roq deletion strain chromatograms of P. chrysogenum were further analyzed for the presence of 13. The chromatogram of the ion with m/z 404.1706, representing the protonated molecule [MϩH] ϩ with formula C 22 H 22 N 5 O 3 of both compound 6 and compound 13, was extracted in a 5-ppm mass accuracy window.…”
