Production of Manganese Peroxidase by Trametes villosa on Unexpensive Substrate and Its Application in the Removal of Lignin from Agricultural Wastes

Abstract: Manganese peroxidase (MnP) is a ligninolytic enzyme that is involved in the removal of lignin from the cell wall of plants. This removal facilitates the access of hydrolytic enzymes to the carbohydrate polymers that are hydrolyzed to simple sugars, which allows the subsequent fermentation to obtain bioproducts, such as ethanol. In this work, response surface methodology (RSM) was employed to optimize the culture conditions on unexpensive substrate for MnP secretion by Trametes villosa. Three independent variab… Show more

“…In the LiP activity assay, an initial analysis was performed without H 2 O 2 , followed by a subsequent analysis with H 2 O 2 after 5 min. All assays with fungal enzyme extracts were performed in triplicates and compared with the corresponding boiled fungal extract, as previously described [35]. The resulting LiP and MnP activities (expressed in U/g) were subjected to analysis using two-way ANOVA followed by the Bonferroni test at 95% confidence.…”

“…Trametes villosa (Sw.) Kreisel, a tropical species that is commonly found in the semiarid region of Brazil [31], is promising for its production of ligninolytic enzymes. There are several works about its application in the decomposition of lignin related to the expression and secretion of its laccase (Lac) enzymes [32][33][34] and MnP's [35][36][37], but there have been no reports on the identification and characterization of LiP, an unusual enzyme detected in most white-rot Basidiomycota that has a central role in lignin degradation [26,29].…”

“…In the present study, optimum enzyme production was tested under the design of RSM with central composite design (CCD) using DOE 8.0.7.1 trial version software (Silva et al, 2014). Banana stalk that resulted in the highest production of lignocellulolytic enzymes was solely optimised through RSM.…”

“…The recalcitrance of lignin against biodegradation is due to its bulky and aromatic nature that limits the accessibility of microbial enzymes and decreases its biodegradability. Therefore, the degradation of lignin is the key step to lignocellulose transformation (Silva et al, 2014).…”
