Preventing Varicella-Zoster Disease

Abstract: Varicella-zoster virus (VZV), the cause of chickenpox and shingles, is a pathogen in retreat following the introduction of mass vaccination in the United States in 1995. The live attenuated Oka vaccine, which is safe and immunogenic, gives good protection against both varicella and zoster in the short to medium term. It has undoubtedly been highly effective to date in reducing all forms of varicella, especially severe disease. However, the huge pool of latent wild-type virus in the population represents a cont… Show more

“…The efficacy rates varied from 85% 30,50 to 100% 27 . The lowest effectiveness (85%) reflected the use of results from studies following introduction of the vaccine in the United States 55,56 , while the most optimistic used data from an older clinical trial 13 . In studies that used the MMRV vaccine, Hammerschmidt et al 52 assumed a 95% efficacy for two doses of vaccine, based on the opinion of experts.…”

“…The smaller decrease in genome copies than virus titers may indicate that not all viral DNA detected in ganglia represents infectious virus. We do not consider infectious KOS-gDA3C virus in the DRG a contraindication for pursuing gD entry-impaired virus as a vaccine candidate since live attenuated varicella zoster virus vaccine is safe and highly effective in humans despite vaccine virus infecting the ganglia [51].…”
