In this study, of 124 samples of ground beef and chicken meat, 101 (81.5%) were positive for Listeria spp. Listeria innocua (35.5%) and Listeria monocytogenes (26.6%) were the most frequently isolated species. All 33 (100%) L. monocytogenes isolates were positive for the virulence genes hlyA, actA, inlA, inlB, inlC, inlJ, prfA, plcA, and iap. Serotype 1/2a, which is one of the listeriosis‐associated serotypes, was the most predominant serotype (57.6%), followed by 1/2c (39.4%) and 3c (3%). In polymerase chain reaction‐restriction fragment length polymorphism (PCR‐RFLP) analysis of inlA for subtyping of the L. monocytogenes strains, the AluI and MluCI enzymes generated two different profiles. Almost all chicken strains were grouped in similar PCR‐RFLP profiles with both restriction enzymes. Most Listeria isolates were resistant to ceftriaxone (79.7%), fusidic acid (63.3%), and clindamycin (47.7%). Resistance to penicillin and ampicillin occurred in 1.6 and 17.9% of the isolates, respectively. The percentage of resistance to three or more agents was 48.1%. Practical application The presence of Listeria spp., especially L. monocytogenes, in foods is inevitable because of the ubiquitous nature of Listeria spp. Illnesses resulting from the consumption of contaminated food severely affect sensitive groups, especially pregnant women, unborn fetuses and newborns as a result of transmission across the placenta or during delivery. Immunosuppressed adults are also sensitive to infection. Listeria grows well at cold temperatures, so storage of contaminated food in the refrigerator can also increase the risk of infections for consumers. In particular, limiting the exposure of pregnant women and immunosuppressed patients to potential sources of Listeria such as raw and undercooked meats is recommended.

Listeria monocytogenes is among the most important foodborne pathogens. It may enter foodprocessing environments through raw materials, handlers or equipment and may persist due to ineffective cleaning or sanitation. The bacterium can be isolated from both frozen vegetables and fresh food substances. This study aimed to estimate the prevalence of L. monocytogenes in spices and frozen vegetables and screen for some virulence factors and drug-resistance determinants of the isolated bacteria. First, conventional microbiological methods were used for the isolation and identification of bacteria. Next, the identity of isolated bacteria was confirmed by molecular techniques, and the virulence genes iap and hlyA were identified by real-time polymerase chain reaction (PCR). The hemolytic activity of the isolates was assessed by cultivation on sheep blood agar. Furthermore, the antimicrobial susceptibility of confirmed L. monocytogenes isolates was tested by the disk diffusion method against 10 antibiotics. Out of 331 vegetable samples, 47 isolates were confirmed to contain L. monocytogenes, whereas none of 40 spice samples tested positive. All isolates were positive for iap and hlyA genes. Susceptibility testing indicated that all isolates were sensitive to trimethoprim/ sulfamethoxazole, but only 36% were sensitive to penicillin G, while 100% and 70% showed intermediate resistance to chloramphenicol and erythromycin, respectively. All tested isolates were resistant to amoxicillin, gentamicin and norfloxacin; on the other hand, 90, 86 and 84% of the tested strains were resistant to ciprofloxacin, ceftazidime/clavulanic acid and amikacin, respectively. In summary, L. monocytogenes isolates disseminated in frozen vegetable samples from the Egyptian market were highly virulent, entirely multiple-drug resistant and were enriched in iron-containing vegetables. Since L. monocytogenes is primarily pathogenic to humans and causes a life-threatening disease, there is a potential infection risk for people who usually deal with frozen vegetables before cooking. Hence, surveillance to L. monocytogenes in frozen products, together with implementation of tight measures would be valuable in preventing listeriosis, and are highly recommended.

BACKGROUND: Some routes of transmission of Escherichia coli O157:H7 to fresh produce include contaminated irrigation water and manure polluted soils. The aim of the present study was to determine the genetic relationships of E. coli O157:H7 isolated from some produce growing region in Nigeria using enterobacterial repetitive intergenic consensus (ERIC) DNA fingerprinting analysis. A total of 440 samples comprising leafy greens, irrigation water, manure and soil were obtained from vegetable producing regions in Kano and Plateau States, Nigeria. Genes coding for the quinolone resistance-determinant (gyrA) and plasmid (pCT) coding for multidrug resistance (MDR) were determined using polymerase chain reaction (PCR) in 16 isolates that showed MDR.
