In this paper, novel drug delivery systems (DDS) were designed based on graphene oxide (GO) as nanocarrier, loaded with two natural substances (quercetin (Qu) and juglone (Ju)) at different concentrations. The chemical structure and morphology of the synthesized GO-based materials were characterized by Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), and Raman spectroscopy. The antibacterial activity was evaluated against standard strains, Staphylococcus aureus ATCC 6538, Escherichia coli ATCC 8739, and Candida albicans ATCC 10231. Results demonstrated excellent antimicrobial activity, with a 5 log reduction of E. coli and a 1 log to 3.04 log reduction of S. aureus populations. Reduction rates were above 90%. Biocompatibility tests were also performed on GO-based materials, and the results showed biocompatible behavior for both L929 fibroblast cell line and BT474 breast cancer cells at lower concentrations. The identity of Qu and Ju was demonstrated by matrix-assisted laser desorption/ionization (MALDI) analysis, showing the compounds’ mass with high accuracy. In addition, specific properties of GO made it a versatile matrix for the MALDI analysis. The results of this study indicated that GO-based platforms may be suitable for applications in many areas for the effective and beneficial use of hydrophobic compounds such as Ju and Qu.

Article informationBackground: Infections caused by Extended Spectrum Beta Lactamase-producing wound pathogens are difficult to be eradicated worldwide. The prevalence rates were reported as worldwide rising, Therefore, Imperative work is needed to detect safe, non-toxic and new-effective Therapy. The Aim of The Work:To detect the first gene was isolated from female patient called temonira-1 gene, bearing Extended Spectrum beta Lactamase-producing strains among multi-drug resistant bacteria isolated from patients with wound infections. And to evaluate the antibacterial effects of PROBIOTICS' culture extracts on these isolates.Patients and Methods: Overall, 320 patients selected from those with wound infections seen in Egypt, The Wound swab samples were collected, transported in brain heart infusion broth, isolated in pure culture and bacterial species were identified by the conventional microbiological methods. Antimicrobial susceptibility testing were done, followed by Phenotypic Detection of Extended Spectrum Beta Lactamase production using the double disk synergy test, PCR technique was done to detect isolates bearing the gene. The detected isolates had tested by the PROBIOTICS' culture extracts, using the well-agar diffusion method and the reducing colony count activity technique.Results: [48.54%] of the total multi-drug resistant pathogens identified were positive for ESβL by double disk synergy test. Among 100 ESβL-producers screened, 32 isolates were detected bearing TEM-1 gene that coding for these enzymes by using PCR technique. All of the studied ESβL producers, bearing TEM-1 gene were susceptible to PROBIOTICS' CULTURE EXTRACTS with inhibition zones diameter average inbetween 12 and 21 mm., in addition, [65.6% of strains] showing reducing colony count activity by 100 % [zero CFU]. Conclusion: PROBIOTICS' CULTURE EXTRACTS revealed excellent activity againstESβL-producing microorganisms that bearing TEM-1 gene. So, these extracts may be considered safe therapeutic promising options to treat such infections.

La resistencia antimicrobiana causada por miembros de la familia Enterobacteriaceae productoras de β-lactamasas de espectro extendido (BLEE) constituyen un problema de salud pública a nivel mundial. Las BLEE son enzimas que confieren resistencia a penicilinas y cefalosporinas de primera, segunda, tercera y cuarta generación, mismas que se expresan en mayor proporción en bacterias gramnegativas. Objetivo: Caracterizar enterobacterias productoras de BLEE en hemocultivos de pacientes atendidos en diferentes áreas del Hospital General Isidro Ayora de Loja, periodo 2019-2020. Materiales y métodos: Estudio documental cuantitativo, descriptivo, transversal. La muestra estuvo constituida por 114 aislamientos. Resultados: Las variables más frecuentes del estudio fueron, sexo femenino (54,4%), grupo etario adulto (39,5%) con edad media de 45 años. Las especies bacterianas prevalentes fueron Klebsiella spp., (43,9%) y Escherichia. coli (39,5%), seguidas de Enterobacter spp., (6,1%), Salmonella spp., (3,5%), Citrobacter freundii (2,6%), Proteus mirabilis (1,8%), Morganella morganii (1,8%) y Serratia marcescens (0,9%). Según la susceptibilidad antimicrobiana, existió alta resistencia a todos los betalactámicos probados, mientras que a los aminoglucósidos, fluoroquinolonas y carbapenémicos presentaron sensibilidad. Las especies predominantes fueron Klebsiella spp., en Hospitalización y Escherichia coli en Emergencia. De todas las especies Klebsiella spp., y Escherichia coli son BLEE (32,5%). Conclusiones: La resistencia a los antimicrobianos causada por Enterobacterias productoras de BLEE se considera un problema de salud grave. Por ello, es importante evaluar y establecer medidas de prevención, control y difusión de las BLEE para reducir la farmacorresistencia y proporcionar un tratamiento eficaz.
