Preparation and Characterization of Thermally Stable Collagens from the Scales of Lizardfish (Synodus macrops)

Abstract: Marine collagen is gaining vast interest because of its high biocompatibility and lack of religious and social restrictions compared with collagen from terrestrial sources. In this study, lizardfish (Synodus macrops) scales were used to isolate acid-soluble collagen (ASC) and pepsin-soluble collagen (PSC). Both ASC and PSC were identified as type I collagen with intact triple-helix structures by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and spectroscopy. The ASC and PSC had high amino acids of … Show more

“…Relative to dry swim bladder weight, the respective yields of ASC and PSC were 33.38% and 84.79%, suggesting that pepsin was able to markedly enhance extraction efficiency, in line with prior reports [ 15 , 16 , 17 ]. This is at least partially attributable to the covalent telopeptide crosslinking of collagen molecules, as pepsin can cleave telopeptide residues to enhance overall collagen solubility [ 18 ].…”

“…38 Further, the solubility of the samples gradually increased with an increase in pH from 7 to 11 (p < 0.001) due to the net negative charge on protein or peptides caused by the dif ference in pH and pI values. 39 Although the solubility pattern of P coll. and PSCH is similar, it is slightly higher for PSCH than P coll.…”

“…Wang et al separated types I and V collagens from the skin of deep-sea redfish by chromatographic techniques [ 7 ]. Chen et al separated type I collagen from the scales of the lizardfish and maintained the triple-helical structures with no cytotoxicity [ 8 ]. Recently, some Japanese scholars extracted collagen from the scales of barramundi ( Lates calcarifer ), which was found to be comparable to that of mammals and showed potential for three-dimensional cell cultivation [ 9 ].…”
