Preparation and characterization of an aptamer-functionalized solid-phase microextraction fiber and its application in the selective monitoring of adenosine phosphates with liquid chromatography and tandem mass spectrometry

Abstract: An aptamer with adenosine triphosphate as a ligand was immobilized onto the surface of a porous-polymer-coated fiber to obtain an aptamer-functionalized porous-polymer-coated solid-phase microextraction fiber. The fiber was observed with a crosslinked and porous morphological surface structure. It shows specific selectivity to adenosine triphosphate with a selectivity coefficient of 22.1 compared to the scrambled oligonucleotide functionalized fiber, and the selectivity factors over other analogues and referen… Show more

“…Aptamer, which is selected through an in vitro selection process called selective evolution of ligands by exponential enrichment (SELEX), are oligonucleotide or peptide molecule that can bind to a specific target molecule , exhibiting high selectivity, affinity, and specificity, equal to or sometimes even better than those of antibodies . This unique property has been widely used in biosensing applications, including detection based on fluorescence intensity, electrochemistry, or color change .…”

“…The sample preparation is essential before HPLC analyses for removing impurities and concentrating the analytes of the interest . Over the years, several techniques have been used for the sample preparation including a wide range of applications of SPE . The latest development in the field of sample preparation involves miniaturization of the techniques.…”

“…A 25 mg sample of the synthesized polymeric material was packed into an empty cartridge column (1 mL). An equal volume of 0.1 M phosphate buffer solution (pH 7.0) was added with known amount of ATP stock solutions (10 and 100 µM) and then passed through the solid phase at a flow rate of about 1.0 mL/min for 10 min by a vacuum system . The collected fractions were filtered through a 0.45‐µm MCE membrane filter unit, and analysis of the ATP template was determined using the HPLC method.…”
