In this study, we aimed to find out new herbal materials that are able to inhibit the growth of the P. aeruginosa and E.coli clinical isolates that has antibiotic resistance. Clinical isolates used in this research are E. coli (n=1) and P.aeruginosa (n=1). Antibiotic susceptibility profiles of E. coli and P. aeruginosa were determined using e-test. Plants were collected in Trabzon region of Turkey are Calendula officinalis, Hypericum perforatum and Glycyrrhiza glabra. DMSO were used as solvent and solid-liquid extraction was employed. Micro-dilution method was preferred fo the determination of the minimum inhibitory concentration (MIC). MIC results were obtained through observation of turbidities. According to E-test results, while P. aeruginosa was resistant to piperacillin, piperacillin/tazobactam, meropenem and ceftazidime, E. coli was resistant to piperacillin, cefotaxime and ceftazidime. DMSO extract of Calendula officinalis showed very strong activity against PA1 with the best MIC (5 mg/mL). DMSO extract of three plant had lower MIC values (5-10 mg/ml) for EC1 and PA1 than ampicillin. In future studies antibacterial activity of different solvents extracts of these plants and other plants against antibiotic resistant clinical isolates will be examined. Natural products from plants are promising in fighting with antibiotic-resistant bacteria.

Focusing on its application in reproductive biotechnology, we evaluated the effects of the essential oil of Syzygium aromaticum (EOSA) on bovine epididymal sperm quality variables, including morphology, membrane functional integrity, membrane structural integrity, mitochondrial activity, metabolic activity, motility and oxidative stress by reactive oxygen species (ROS) levels. Bovine spermatozoa from eight males were incubated into the following groups: EOSA0 (without EOSA), EOSA10 (10 μg/ml of EOSA), EOSA15 (15 μg/ml of EOSA) and EOSA20 (20 μg/ml of EOSA); the incubation time with and without the EOSA was 1 or 6 hr. None of the sperm quality variables presented difference among the EOSA concentrations. However, the incubation time had a significant effect on the membrane functional integrity, membrane structural integrity, mitochondrial activity, progressive motility and some kinetic parameters. The effect of interaction among EOSA and incubation time was significant only on ROS levels. Spermatozoa incubated in the presence of 15 μg/ml of the EOSA for 1 hr had significantly reduced ROS levels compared with all other groups in the same time. In conclusion, the EOSA at a concentration of 15 µg/ml has antioxidant effects and protects bovine epididymal spermatozoa; hence, the EOSA may potentially be used in the field of reproductive biotechnology.

The Helicobacter pylori bacterium is one of the main causes of chronic gastritis, peptic ulcers, and even gastric cancer. It affects an average of half of the world population. Its difficult eradication depends upon multi-drug therapy. Since its classification as a group 1 carcinogenic by International Agency for Research on Cancer (IARC), the importance of H. pylori eradication has obtained a novel meaning. There is considerable interest in alternative therapies for the eradication of H. pylori using compounds from a wide range of natural products. In the present study, we investigated the antibacterial property of the isocoumarin paepalantine against H. pylori and it exhibited significant anti-H. pylori activity at a minimum inhibitory concentration (MIC) of 128 μg/mL and at a minimum bactericidal concentration (MBC) of 256 μg/mL. The scanning electron microscopy (SEM) revealed significant morphological changes of the bacterial cell as a response to a sub-MIC of paepalantine, suggesting a penicillin-binding protein (PBP) inhibition. Computational studies were carried out in order to study binding modes for paepalantine in PBP binding sites, exploring the active and allosteric sites. The data from the present study indicates that paepalantine exhibits significant anti-H. pylori activity, most likely by inhibiting membrane protein synthesis.
