Polypeptide profiles of South Indian isolate of Trypanosoma evansi

Abstract: The field isolates of Trypanosoma evansi was collected from the infected cattle and it was propagated in rats. Trypanosoma evansi parasites were separated from the blood of infected rats by using diethylaminoethyl cellulose column chromatography. Whole cell lysate antigen (WCL) was prepared from purified trypanosomes by ultrasonication and centrifugation. The prepared WCL antigen was further purified by 50 % ammonium sulphate precipitation. Protein concentration of WCL antigen of T. evansi was 60 mg/ml. Protei… Show more

“…Previously 8 polypeptide bands size of 25, 34, 37, 42, 53, 60, 74 and 85 kDa recorded from isolate of cattle. Similarities between the cattle isolate and present isolate was noticed at the band size of 34, 53 and 64 kDa molecular sizes [2].…”

“…The approximate molecular weights of these polypeptides were ranged from 34 to 98 kDa with relative molecular weight of 34, 48, 53, 55, 64, 74, 80 and 98 kDa. Whole cell lysate antigen of T. evansi isolate was prepared according to the previous studies [2]. Recorded polypeptide profiles of diminazene aceturate resistant isolates of T. evansi revealed the presence of different bands from the previous studies which indicates the presence of variations in the isolates.…”

“…This characteristic reinforces the importance of the host's humoral immune response and suggests the possibility of controlling this enzootic disease through immunoprophylactic methods. 13 Whole cell lysate (WCL) antigen of T. Evansi isolate was prepared following the method of Sivajothi et al, 14 The approximate molecular weights of these polypeptides were ranged from 25 to 82kDa. 14 Hence, the present findings were compared with the polypeptide profiles of the cattle isolates of T. Evansi which revealed the presence of additional bands indicating the variations in the isolates between the different hosts.…”

“…Crude sonicated antigens of T. evansi isolates from different host origins revealed a similar polypeptide profile, and whole cell lysate (WCL) antigens prepared from T. evansi isolated from a unique host might be useful for serodiagnosis of trypanosomiasis in several species of animals ( Laha & Sasmal, 2008 ; Sivajothi et al., 2016 ). High levels of specificity for serum agglutination tests can be obtained by acidic buffering of the antigen (pH ∼3.65), which decreases the activity of agglutinins with less specificity, such as IgM, thus favoring reactions based on IgG ( Corbel, 1972 ; Patterson et al., 1976 ).…”
