Plasminogen Activator in Bronchoalveolar Fluid

Abstract: Plasminogen activator was purified from the soluble fraction of bronchoalveolar lavage fluid, and biological and immunological characteristics of the activator were examined by electrophoretic enzymography. The purified fraction showed a single band with a molecular weight of 53,000. The enzyme activity was eliminated in the presence of DFP and did not display fibrin affinity. Immunological tests revealed that the plasminogen activator reacted with IgG of antiurokinase but not with that of tissue-type plasmino… Show more

“…8 Plasminogen activator activity was measured by the fibrin film method 9 as well as by electrophoretic enzymography. 10 Electrophoretic enzymography was performed with sodium dodecyl sulfatepolyacrylamide gel 11 containing plasminogen-rich fibrin. After electrophoresis, the gel was placed in 2.5% Triton X-100 and washed three times.…”

“…The assay of u-PA and t-PA activity was performed as previously described [11]. Briefly, 10% polyacrylamide gel contained 0.55 mg/ml bovine plasminogen-rich fibrinogen (Organon Teknika; Dublin, Ireland) and 0.056 NIH U/ml thrombin (Sigma-Aldrich; St. Louis, MO, USA).…”
