Plaque Formation by Mumps Virus and Inhibition by Antiserum

Abstract: Boston and ABC strains of mumps virus produced plaques approximately 1.0 mm in diameter in monolayers of BGM cells. The plaques were circular and either clear or target-like in form. Ricki strain virus produced plaques of similar size and form but, in addition, a red plaque was observed with this agent. The vaccine strain of mumps virus, Jeryl Lynn, produced minute clear plaques approximately 0.3 mm in diameter. Incorporation of diethylaminoethyl (DEAE)-dextran into the overlay medium did not affect the size d… Show more

“…HAI and neutralization tests showed the presence of antibody in serum samples. The magnitude of response was significant and similar in most cases to human cases of natural mumps (7). The lowest antibody response observed was that occurring in the animal which received a 25-mg dose of cortisone acetate.…”

“…This agent was passaged nine times in primary human amnion cell cultures and two or three times in primary rhesus thyroid cell cultures. The preparations used to inoculate animals titered 4 X 107 plaque-forming units (PFU)/ml (7). Virus diluent was medium 199 with 2% newborn calf serum.…”

“…Infectivity assays. Infectious measles virus and mumps virus were assayed by a plaque formation method in BGM cells as previously described for mumps virus (3). VSV was also titered by plaque assay.…”

“…Virus antibody. Antibody to mumps virus was determined by a plaque reduction neutralization test (7). The titers were expressed as the reciprocal of the dilution of serum or parotid extract which resulted in a 50O reduction of input virus.…”
