Euphorbia fischeriana has a long-standing history of use in traditional medicine for the treatment of tuberculosis diseases. However, the plant’s therapeutic potential extends beyond this specific ailment. The present study aimed to investigate the antioxidant properties of Euphorbia fischeriana and lay the groundwork for further research on its potential therapeutic applications. Phytochemical tests were performed on the plant, and 11 types of phytochemicals were identified. Ultraviolet–visible spectrophotometry was used to evaluate the active components and antioxidant properties of eight different solvent extracts, ultimately selecting acetone extract for further research. UHPLC-ESI-Q-TOF-MS identified 43 compounds in the acetone extract, and chemical calculations were used to isolate those with high content and antioxidant activity. Three stability experiments confirmed the extract’s stability, while cell viability and oral acute toxicity studies demonstrated its relatively low toxicity. In rats, the acetone extract showed significant protective effects against D-galactosamine-induced liver damage through histopathological examination and biochemical analysis. These results suggest that Euphorbia fischeriana’s acetone extract has potential in treating diseases related to oxidative imbalances. Therefore, this study highlights the plant’s potential therapeutic applications while providing insight into its antioxidant properties.

The main objective is to determine the most suitable extraction technique for maximizing alkaloid yield and unlocking their potent biological activities. Extraction parameters, including yield, antioxidant activity, and anti-inflammatory activity, will be meticulously assessed. This study's findings will contribute to understanding the alkaloid profile of Marrubium vulgare L leaves and facilitate the development of effective extraction techniques for obtaining high-quality alkaloidrich extracts. Material and Method Chemicals and plant materialThe glassware used was purchased from Pyrex. Solvents (methanol, ethanol, hexane, chloroform, ethyl acetate, and diethyl ether) were purchased from Honeywell Fluka. Chemicals and reagents (Mayer's reagent, iron chloride, sodium hydroxide, ammonia, hydrochloric acid, sulfuric acid, sodium bicarbonate, sodium phosphate, ammonium molybdate, ammonium hydroxide, potassium ferricyanide, trichloroacetic acid, ascorbic acid, lithium chloride, Gel filtration for chromatography Sephadex G-50, aspirin, and formol) were purchased from Sigma-Aldrich (Casablanca, Morocco). Extracts concentration was done using a rotary evaporator (BUCHI, Rotavapor R-100, Germany), and optical density measurements were performed on Ultraviolet/Visible spectrophotometer (Shimadzu UV-1600 PC UV, Japan). Molecular structures and physicochemical data were obtained from PubChem and SwissADME programs (https://pubchem.ncbi.nlm.nih.gov/, http://www.swissadme.ch/). Plant material used consists of Marrubium vulgare L leaves, which were collected during February and March 2021 in the Fez-Meknes region and were identified by Dr. Chaimae Rais in The National Agency for Medicinal and Aromatic Plants, Taounate, Morocco (Document no. 075/2021). The study was conducted at Human Pathology, Biomedicine and Environment Laboratory, in the Faculty of Medicine and Pharmacy (34.00612274141898, -4.964672169974484).

The present work describes the in vitro biological activities of the crude extracts (petroleum ether, ethyl acetate and n-butanol) prepared from the species Ephedra altissima Desf. The estimation of total phenolic, flavonoid and tannin contents were carried out using the Folin-Ciocalteu, trichloroaluminum and acidified vanillin methods, respectively. The evaluation of the in vitro antioxidant activities were performed by three different methods namely: scavenging of the free radical ABTS, permanganate reducing antioxidant capacity, and potentiometric assay. In addition, the antibacterial activity was assessed by the agar disk diffusion assay against seven bacterial strains. The results of the phytochemical screening revealed the presence of several types of secondary metabolites. The EtOAc extract exhibited the highest content of phenols (125.62 ± 1.51 μg EGA mg-1 of extract). The greatest flavonoid and tannin contents were observed for n-BuOH extract (19.18 ± 0.39 µg EQ mg-1 of extract and 8.95 ± 1.70 μg EC mg-1 of extract, respectively). Moreover, the EtOAc extract revealed potent antioxidant activity in all the tested methods. Furthermore, the aqueous extract from the species E. altissima showed a good ability to reduce iron III to iron II with a value of 0.68 ± 0.3 moL eq L-1 in potentiometric assay. All the crude extracts (PE, EtOAc and n-BuOH) displayed inhibition of bacterial growth against at least three strains with values of MIC ranging from 3.125 to 50 µg mL-1. Therefore, these results suggest that Ephedra altissima could be used as an important source of natural bioactive compounds with antioxidant and antibacterial properties.
