Methanol extract of Indigofera hirsuta, was evaluated for its antiradical potential and capacity in inhibiting lipoxygenase and aldose/aldehyde reductase enzymes. The ethyl acetate fraction derived from the methanol extract partition, showed the greatest antioxidant capacity, while the butanol was the strongest inhibitor of lipoxygenase enzyme. All fractions (diethyl ether, ethyl acetate, butanol and the aqueous residue) exhibited strong inhibition capacity of both aldose/aldehyde reductase enzymes, which comes in agreement with the ethnomedicinal plant utilization as an antidiabetic agent.LC-DAD-MS(ESI+) fraction analysis verified the findings above, leading to a conclusion regarding the biological activities attributed to the main compounds.Phytochemical analysis led to the identification of an indolic dimer, cinnamic acids, phenolics, flavonoid glycosides, a cyclic polyol, the rare sugar 1-methyl-β-Dglucopyranoside and glycerol. Many of these compounds were isolated for the first time in Indigofera species while the indolic dimer was isolated for the first time in the Fabaceae family.

Rhoifolin (apigenin‐7‐O‐β‐neohesperidoside) belongs to the class of flavonoids and was reported to exhibit anti‐inflammatory, cytotoxic, antidiabetic, hepatoprotective, and cardioprotective activities. The current study presents the in‐vitro evaluation of the antioxidative effects of rhoifolin by many assays, namely DPPH, CUPRAC, ABTS, phosphomolybdenum, and FRAP. Enzyme inhibitory potential was also evaluated for acetylcholinesterase (AChE), butyrylcholinesterase (BChE), tyrosinase, amylase, and glucosidase enzymes. While results revealed weak antioxidant activities for rhoifolin, the compound demonstrated some promising enzyme inhibitory effects against BChE (4.03 mg GALAE/g) and tyrosinase (7.44 mg KAE/g) but was not active on AChE. Regarding anti‐diabetic enzymes, the compound was active on amylase but did not show any inhibition effect on glucosidase. In‐silico molecular docking study was performed for rhoifolin on the active site of NADPH oxidase, BChE, and amylase enzymes to verify the observed enzyme inhibitory effect. Good binding affinities were observed for rhoifolin on all the docked enzymes, revealing numerous hydrogen bonds, carbon‐hydrogen, van der Waals interactions. This is the first study to evaluate the enzyme inhibition potential of rhoifolin. We concluded that the increase in the degree of glycosylation might decrease the antioxidant abilities of flavonoids and that rhoifolin had moderate enzyme inhibition abilities to be investigated in future studies.

The inhibitory activities of Matricaria recutita L. 70% methanol extract were evaluated by isolating and testing 10 of its compounds on rat lens aldose reductase (RLAR), advanced glycation end products (AGEs), and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging. Among these compounds, apigenin-7-O-β-D-glucoside, luteolin-7-O-β-D-glucoside, apigenin-7-O-β-D-glucuronide, luteolin-7-O-β-D-glucuronide, 3,5-O-di-caffeoylquinic acid, apigenin, and luteolin showed potent inhibition, and their IC50 values in RLAR were 4.25, 1.12, 1.16, 0.85, 0.72, 1.72, and 1.42 μM, respectively. Furthermore, these compounds suppressed sorbitol accumulation in rat lens under high-glucose conditions, demonstrating their potential to prevent sorbitol accumulation ex vivo. Notably, luteolin-7-O-β-D-glucuronide and luteolin showed antioxidative as well as AGE-inhibitory activities (IC50 values of these compounds in AGEs were 3.39 and 6.01 μM). These results suggest that the M. recutita extract and its constituents may be promising agents for use in the prevention or treatment of diabetic complications.
