Pharmacokinetic study of metoprolol in rabbit plasma by capillary electrophoresis with laser-induced Fluorescence detection

Abstract: This work described a sensitive method for determination of metoprolol in rabbit plasma. The method involved purification by ultrafiltration, derivatization with Fluorescein isothiocyanate, separation by capillary electrophoresis and determination by laser induced fluorescence detector. Other components in plasma including a variety of amino acids and proteins did not interfere with the determination of metoprolol under experimental conditions. The assay had a wide range (2.0-500 ng/mL) of linearity and a dete… Show more

“…The results of the analysis were compared with those from other methods. The LODs and LOQs of Sot and Met for standard samples (or urine samples) were lower than those obtained via colorimetry [4], spectrophotometry [5], ECD [6,7], HPLC‐photodiode array UV [14], HPLC‐ECL [15], CE‐UV [16], CE‐ECD [17], and CE‐ECL [18], but higher than those obtained via GC‐MS [8], HPLC‐MS/MS [9,10] HPLC‐fluorescence [12], HPLC‐UV [13], and CE‐LIF [19]. Wide linear ranges were obtained using most methods.…”

“…These include colorimetry [4], spectrophotometry [5], electrochemical detection (ECD) [6,7], GC‐MS [8], HPLC methods with MS/MS [9–11], fluorescence [12], UV [13,14], and electrochemiluminescence (ECL) detection [15]. Additionally, CE‐UV [16], CE‐ECD [17], CE‐ECL [18], and CE‐LIF detection [19] has been established.…”

“…AT undergoes little or no metabolism in the liver and is mainly excreted in urine (over 85% of the absorbed drug is excreted in urine as unchanged drug) [1]. Different analytical techniques have been reported for the analysis of AT such as high performance liquid chromatography (HPLC) [2][3][4], spectrophotometric [2,5,6], capillary electrophoresis [7,8], gas chromatography (GC) [9], and spectrofluorimetric [10] methods. Paracetamol (PR) or acetaminophen (Figure 1(b)) is an over-the-counter (OTC) analgesic and antipyretic drug.…”
