Overexpression of lncRNA H19 enhances carcinogenesis and metastasis of gastric cancer

Abstract: Long non-coding RNAs (lncRNAs) play key roles in the progression and metastasis of some carcinomas. We previously showed that the expression of lncRNA H19 (H19) was higher in gastric cancer (GC) tissues than that in paired noncanerous tissues. However, the underlying mechanisms remain unclear. In this study, H19/miR-675 knockdown models in the MKN45 cell line and ectopic expression models in the SGC7901 cell line were established, and a co-expression network of H19 was generated to identify target genes by RIP … Show more

“…The results suggested that 14 lncRNAs were associated with the above indicators, such as HOTAIR, TINCR and H19. These indicators of lncRNAs mainly focus on tumor grade, TNM staging and lymphatic metastasis, among these lncRNAs, H19, and HOTAIR also were reported to be indicators of invasion and lymphatic metastasis of GC (56,57). However, the function of other lncRNAs has not been reported with relevant features yet.…”

“…Using high-throughput technologies, thousands of lncRNAs have recently been identified, and a large number of studies have demonstrated that dysregulated lncRNAs are closely related to human cancers [7], such as HCC [8], lung cancer [9], breast cancer [10], and colorectal cancer [7]. The expression levels of certain lncRNAs are associated with the recurrence [11], metastasis [12], and prognosis of cancers. Therefore, the identification of cancer-associated lncRNAs will lead to a better understanding of the molecular biology of tumors and aid in developing novel therapeutic targets.…”

“…H19 is a member of a highly-conserved cluster of imprinted genes, and was the first lncRNA to be associated with cancer (12). Previous studies have indicated that H19 exhibits dysregulated expression in GC (13,14), hepatocellular carcinoma (15), bladder cancer (16), breast cancer (17) and esophageal cancer (18), and that itmay cause tumor promoting and suppressing effects (19). Niu et al (20) screened lncRNA expression profilesfrom colorectal cancer and adenoma samples, and identified a novel lncRNA, AK027294, which was associated with cell proliferation, migration and apoptosis in colorectal cancer cells.…”
