Original Nerve Growth Factor Mimetic Dipeptide GK-2 Restores Impaired Cognitive Functions in Rat Models of Alzheimer’s Disease

Abstract: Dipeptide mimetic of the nerve growth factor (NGF) loop 4, hexamethylenediamide bis-(N-monosuccinyl- glutamyl-lysine) (GK-2), was synthesized at the V.V. Zakusov Scientific Research Institute of Pharmacology of the Russian Academy of Medical Sciences. GK-2 exhibited in vitro neuroprotective activity at nanomolar concentrations, was efficient in animal models of the Parkinson’s disease, ischemic and hemorrhagic stroke, and global cerebral ischemia at doses of 0.01–5 mg/kg (intraperitoneally) and 10 mg/kg (per o… Show more

“…To date, there are only a few known NGF mimetics such as GK-2 h, which in experimental models show capacity to promote neuronal survival, differentiation and synaptic plasticity demonstrating possible application for Alzheimer’s disease [27], Parkinson’s disease [28], cerebral ischemia [29], neural toxicity [30] or SCI [31]. The aim of the current investigation is to screen a large variety of natural plant extracts and polyphenolics, in an exploratory fashion, to elucidate substances that exert NGF mimetic effects in rat dopaminergic pheochromocytoma PC-12 cells on collagen coated plates.…”

“…The streptozotocin-treated were significantly impaired as compared to sham rats operated in learning which was measured by a latency to locate of a submerged platform across days of training. GK-2 significantly reduced these spatial learning impairments [33].…”

“…Fimbria/fornix lesion caused significant habituation deficits, which manifested in almost unchanged level of horizontal motor activity. The habituation behavior in GK-2-treated rats was restored [33].…”

“…GSB-106, GSB-214 or GK-2 (dissolved in distilled water) were administered ip at doses of 0.1, 0.1 and 1 mg/kg respectively, daily 4 h after MCAO for 7 days. These doses were chosen as the most pharmacologically active based on preliminary in vivo studies [13,17]. The MCAO group was treated with distilled water in the same regime.…”
