Over the years, brown algae bioactive polysaccharides laminarin, alginate and fucoidan have been isolated and used in functional foods, cosmeceutical and pharmaceutical industries. The extraction process of these polysaccharides includes several complex and time-consuming steps and the correct adjustment of extraction parameters (e.g., time, temperature, power, pressure, solvent and sample to solvent ratio) greatly influences the yield, physical, chemical and biochemical properties as well as their biological activities. This review includes the most recent conventional procedures for brown algae polysaccharides extraction along with advanced extraction techniques (microwave-assisted extraction, ultrasound assisted extraction, pressurized liquid extraction and enzymes assisted extraction) which can effectively improve extraction process. The influence of these extraction techniques and their individual parameters on yield, chemical structure and biological activities from the most current literature is discussed, along with their potential for commercial applications as bioactive compounds and drug delivery systems.(APS) is determined by algae species however it is also influenced by other factors causing inter-species variation, e.g., growth location and harvesting season [8]. Vast structural variation between the APS therefore presents a challenge in terms of pre-treatments application, extraction techniques and optimization, characterization of isolated fractions and determination of their biological properties.Chemical structure and yield of APS isolated from marine macroalgae by conventional extraction (CE) techniques can be affected by various experimental conditions (pH, time, temperature, pressure, particle size, solvent, sample to solvent ratio, agitation speed etc.). In addition, different advanced techniques such as microwave assisted extraction (MAE), ultrasound assisted extraction (UAE), pressurized liquid extraction (PLE), enzyme-assisted extractions (EAE) are assessed and applied for APS extraction [9][10][11].In general, the chemical structure of polysaccharides determines its physical, chemical and biochemical properties as well as its biological activities [12]. Several studies have reported that their biological activity is strongly associated with their chemical structure [9]. Due to very complex mechanisms that are affected by many factors, the correlation between polysaccharide structure and biological activity is still not sufficiently clarified.In order to improve isolation of APS, pre-treatments are usually applied to the algal biomass prior to the extraction process with the two aims: (i) to prevent co-extraction of interfering bioactive compounds with similar solubility; and (ii) to disrupt cell walls and improve mass transfer of APS into extraction solvent. Sericin and alginate particles, prepared by ionic gelation technique, demonstrated to be a promising matrix to controlled release of diclofenac sodium. Ten particle compositions of sericin, alginate, and diclofenac were evaluated. The drug incorporation was confirmed by scanning electron microscopy, Fourier Transform Infrared Spectroscopy, and X‐ray diffraction analysis. In vitro dissolution profile was performed to obtain the drug release profile in gastric and enteric medium. The drug release profile indicated that sericin delays the release and alginate contributes to the gastro‐resistance of formulations. The blend with composition of 2.5% of sericin, 2.8% of alginate with 2.0% w/v of diclofenac demonstrated to be feasible for drug delivery due the entrapment efficiency of 81.06% and release delay of 360 min, the highest time of all investigated compositions. The mathematical modeling showed that the drug release mechanism is associated to the process of swelling, matrix erosion, and a combination of diffusion and chain relaxation mechanisms. © 2017 Wiley Periodicals, Inc. Resumen-Se determinaron las mejores condiciones para la extracción de alginato de sodio a partir del alga parda (Padina durvillaei) y su transformación hidrotérmica a ácido láctico, la cual fue analizada mediante superficie de respuesta BoxBehnken. Las variables evaluadas para la extracción del alginato de sodio a partir del alga fueron: temperatura, tiempo de reacción y concentración alcalina, donde la variable de respuesta fue el rendimiento de alginato. La producción de ácido láctico a partir de alginato y alga se estudió considerando las variables: temperatura, tiempo de reacción y masa de catalizador. La identificación de los grupos funcionales del alginato y la evaluación de la razón ácido manurónico/ácido gulurónico (M/G) se realizó con Espectroscopia Infrarroja de Transformada de Fourier (FTIR). El peso molecular del alginato se determinó con viscosimetría capilar y el comportamiento térmico con Calorimetría Diferencial de Barrido (DSC) y Análisis Termogravimétrico (TGA). El ácido láctico fue cuantificado directamente en el producto final de la reacción por Cromatografía de Gases acoplado a Espectrometría de Movilidad Iónica (GC-IMS). De acuerdo a los resultados obtenidos a partir de la biomasa marina se puede extraer diversos compuestos orgánicos de alto valor agregado, debido a la despolimerización de su estructura en el proceso hidrotermal.Palabras Claves-Algas pardas, alginato de sodio, ácido láctico, superficie de respuesta.Abstract-The best conditions were determined for the extraction of sodium alginate from brown algae (Padina durvillaei) and its hydrothermal transformation to lactic acid, which was analyzed by the Box-Behnken response surface. The variables evaluated for the extraction of sodium alginate from the algae were: temperature, reaction time and alkaline concentration, where the response variable was alginate yield. The production of lactic acid from alginate and algae was studied considering the variables: temperature, reaction time and catalyst mass. The identification of the alginate functional groups and the evaluation of the ratio mannuronic acid / guluronic acid (M / G) was performed with Fourier Transform Infrared Spectroscopy (FTIR). The molecular weight of the alginate was determined with capillary viscosimetry and the thermal behavior with Differential Scanning Calorimetry (DSC) and Thermogravimetric Analysis (TGA). The lactic acid was quantified directly in the final product of the reaction by Gas Chromatography coupled to Ion Mobility Spectrometry (GC-IMS). According to the results obtained from marine biomass, it is possible to extract various organic compounds with high added value, due to the depolymerization of their structure in the hydrothermal process.Keywords-Brown seaweed, sodium alginate, lactic acid, surface response methodology.
