Alginate is a polysaccharide that, for commercial purposes, is extracted exclusively from marine brown algae. In this chapter, we discuss the main sources of alginate and sodium alginate manufacturing, its chemical structure and physicochemical properties, the alginate modifications, and blend formation. We also present applications of alginate and sericin blend in the pharmaceutical and environmental fields as well as case studies.
Sericin and alginate particles, prepared by ionic gelation technique, demonstrated to be a promising matrix to controlled release of diclofenac sodium. Ten particle compositions of sericin, alginate, and diclofenac were evaluated. The drug incorporation was confirmed by scanning electron microscopy, Fourier Transform Infrared Spectroscopy, and X‐ray diffraction analysis. In vitro dissolution profile was performed to obtain the drug release profile in gastric and enteric medium. The drug release profile indicated that sericin delays the release and alginate contributes to the gastro‐resistance of formulations. The blend with composition of 2.5% of sericin, 2.8% of alginate with 2.0% w/v of diclofenac demonstrated to be feasible for drug delivery due the entrapment efficiency of 81.06% and release delay of 360 min, the highest time of all investigated compositions. The mathematical modeling showed that the drug release mechanism is associated to the process of swelling, matrix erosion, and a combination of diffusion and chain relaxation mechanisms. © 2017 Wiley Periodicals, Inc. RESUMO -Este trabalho teve como finalidade estudar a incorporação do fármaco diclofenaco de sódio em uma matriz formada pelos polímeros sericina (obtido do casulo do bicho-da-seda) e alginato (obtido comercialmente). Foram produzidas 3 formulações de partículas, as quais já haviam obtido porcentagens de incorporação altas em estudos anteriores, com diferentes proporções de sericina, alginato e fármaco. Essas partículas foram submetidas a ensaios de liberação em meio gástrico e entérico simulados, apresentando gastrorresistência das partículas e liberação controlada do fármaco devido, principalmente, à presença do alginato e da sericina, respectivamente. Ademais, foram aplicados diferentes modelos matemáticos à curva de liberação em meio tamponado pH 6,8, sendo que o Modelo de Weibull foi o que apresentou melhores resultados (R 2 aj próximo de 1 e menores valores de Critério de Informação de Akaike) em comparação com os demais ajustes. INTRODUÇÃOO casulo do bicho-da-seda é composto por duas principais proteínas: a fibroína e a sericina. A fibroína é utilizada nas indústrias de fiação de seda, enquanto a sericina é tratada como resíduo desse processo. Quando isolada, a sericina apresenta característica de gel; contudo, quando misturada com outros polímeros, obtém-se um material que pode ser aplicado em diversas áreas, como a de cosmético e de fármaco (Aramwit et al., 2012).O alginato é um polissacarídeo derivado das algas marinhas, sendo caracterizado por sua gastrorresistência, o que o torna favorável para a incorporação de substâncias que são sensíveis a esse meio (SINHA et al., 2015). Desta forma, a formação de uma blenda entre a sericina e alginato torna-se interessante, uma vez que resulta em uma matriz adequada para a incorporação de fármacos.O diclofenaco de sódio é um medicamento amplamente utilizado no tratamento de artrite reumatoide e osteoartrite. Por possuir uma meia-vida de 1-2 h, este fármaco requer múltiplas dosagens, causando efeitos adversos como gastrite e úlcera peptídica (Nayak e Pal, 2011). RESUMOO presente estudo visa à avaliação da eficiência de incorporação de diclofenaco de sódio (DS) em partículas de sericina (ser) e alginato (alg) produzidas pelo método de gotejamento seguido de reticulação. A sericina é uma proteína presente nos casulos do bicho-da-seda (Bombyx mori) que apresenta diversas características necessárias para a incorporação de fármacos, sendo que a mistura com o biopolímero alginato pode fornecer características mais adequadas para melhorar a encapsulação. O DS é um antiinflamatório amplamente utilizado e que apresenta efeitos colaterais recorrentes no sistema gastrointestinal, além de necessitar de múltiplas doses diárias para manter o seu efeito terapêutico no plasma sanguíneo. Diante disso, a modificação da forma farmacêutica de apresentação do DS torna-se desejável para minimizar os efeitos colaterais e não adesão do paciente ao tratamento. Formulações com diferentes frações de sericina e alginato em sua composição foram produzidas e avaliadas quanto à eficiência de incorporação de DS, sendo que as partículas F5 (38% ser + 31% alg + 31% DS) e F6 (36% ser + 36% alg + 28% DS) apresentaram-se mais viáveis para a encapsulação, com eficiências de 56,3% e 47,1%, respectivamente. Sendo assim, análises de MEV para verificar a morfologia das superfícies e FTIR para verificar a interação da blenda com o DS, das formulações F5 e F6 e da formulação F12 (sem presença de sericina) foram realizadas, sendo possível comprovar as melhoras das características de encapsulação do DS na presença da blenda de sericina e alginato.
