One hundred and one new simple sequence repeat-based markers for the canine genome

Abstract: One hundred and one new dinucleotide repeat polymorphisms specific for the canine genome have been identified and characterized. Screening of both primary libraries and marker-selected libraries enriched for simple sequence repeats led to the isolation of large numbers of genomic clones that contained (CA)n repeats. Over 200 of these clones were sequenced, and PCR primers that bracket the repeat were developed for those that contained ten or more continuous (CA)n units. This effort led to the production of 101… Show more

“…All samples were profiled at the mitochondrial DNA (mtDNA) control region with primers described in Wilson et al (2000) and 12 autosomal microsatellite markers (cxx225, cxx200, cxx123, cxx377, cxx250, cxx204, cxx172, cxx109, cxx253, cxx442, cxx410 and cxx147; Ostrander et al, 1993Ostrander et al, , 1995 with conditions described in Wheeldon and White (2009). Gender was determined either by amplification at the Zfx/Zfy intron (Shaw et al, 2003) or with Zfx/Sry primer pairs (Aasen and Medrano, 1990;Fain and LeMay, 1995).…”

“…Specifically, the loci that we used were AHT125, PEZ19 and FH2137 [19], FH2054 and FH2226 [20], AHT121 [21], C05.377 [22] and CXX.20 [23]. Polymerase chain reactions (PCRs) for blood and faecal samples were carried out in 10 ml reactions containing 5 ml of Qiagen Multiplex PCR Master Mix, 1 ml Q-Solution and 0.06 mM of FH2054, 0.08 mM of AHT125, 0.…”

“…Based on the criteria, an AHT125 marker [1,6], nine CPH markers [3,4] and six CXX markers [2,8,10,12] were selected. In addition, four CXX markers (CXX419, 453, 618 and 629) were tested in this study, although their He and PIC information was not described previously [11].…”
