RESUMOA leishmaniose visceral canina é uma importante zoonose causadora de sinais clínicos cutâneos e viscerais, que tem o cão como principal reservatório urbano. No Brasil, vários estados são endêmicos para a doença, principalmente no nordeste e norte do país. Com o aumento do deslocamento populacional e de animais de companhia os relatos da doença vêm sendo descritos crescentemente, como é o caso do Paraná que já é considerado endêmico em alguns municípios desde 2013. Descreve-se um caso alóctone de leishmaniose visceral canina diagnosticado em capa leucocitária, com ênfase nas alterações laboratoriais. Um cão da raça Chow-Chow, macho, de 7 anos proveniente de área endêmica (Tocantins) foi atendido em um Hospital-escola no Paraná. O animal apresentava epistaxe, secreção purulenta periocular; mucosas hipocoradas; claudicação, fraqueza muscular, ataxia e cansaço progressivo, abdômen rígido e álgico a palpação. O hemograma revelou intensa pancitopenia (1,68x10 6 eritrócitos/µL; 1.800 leucócitos totais/µL e 29.000 plaquetas/µL) e discreta hipoproteinemia (5,8g/dL) e estruturas compatíveis com Anaplasma sp em plaquetas. Os valores de alanina aminotransferase, fosfatase alcalina, uréia e creatinina estavam dentro do intervalo de referência para a espécie. Foi analisada uma lâmina de capa de leucócitos, na qual foram observadas formas amastigotas de Leishmania sp. em neutrófilos e monócitos. Foram notificados os órgãos responsáveis e o tutor optou pela eutanásia. PALAVRAS-CHAVE: capa leucocitária, leishmaniose visceral canina, pancitopenia

Canine leishmaniasis (CanL) is a zoonotic vector-borne disease that is endemic in the Mediterranean Basin including Morocco. Dogs play a major epidemiological role in this zoonosis as reservoir hosts. This study investigated the clinical manifestations of CanL in dogs naturally infected with Leishmania infantum. A total of 96 dogs presented to the Small Animal Clinic of the Hassan II Agronomy and Veterinary Institute (IAV Hassan II) of Rabat, Morocco, and were tested by RT-PCR and/or serology. Among them, 32 (33.3%) were positive to Leishmania infantum infection. The majority of the positive dogs (93.7%) came from urban areas. Most of them were male (62.5%) and purebreds (65.6%), were aged between 3 and 7 years (71.8%), and had outside activities (guarding, hunting, livestock guarding, and service activities) (71.8%) and all of them were living exclusively outdoor or had free access to the outdoor environment. Lymphadenomegaly (81.2%), dermatological disorders (65.6%) (mostly exfoliative dermatitis), weight loss (59.3%), exercise intolerance (56.2%), anorexia (28.1%), hyporexia (15.6%), and ocular lesions (28.1%) were the most frequent clinical signs and complaints recorded. Anemia and hyperproteinemia due to hyperglobulinemia were observed in 68.7% and 72.7% of the cases, respectively. These results suggest that CanL leads to various nonspecific clinical signs as described previously, making the diagnosis challenging. Since CanL is endemic in Morocco, it should be recommended to systematically test dogs displaying clinical signs compatible with this disease and to regularly screen asymptomatic at-risk dogs. It is also crucial to educate dog owners about the zoonotic aspect of the disease and to encourage intersectorial collaboration following the “One Health” concept, in order to contribute to a more effective control/prevention of human and canine leishmaniasis.

Background Ocular manifestations in dogs with leishmaniasis are frequent and complications in affected tissues can lead to blindness. Immune processes play a very important role in the pathogenesis of ocular inflammation. Therefore, the immunology of ocular manifestations in dogs with leishmaniasis remains complex and poorly understood. Objectives Estimation and characterisation of ocular and periocular manifestations in dogs naturally infected with Leishmania infantum and investigation of the production site of specific anti‐Leishmania infantum IgG. Methods The present investigation used 53 confirmed dogs infected with Leishmania infantum, presenting ocular and periocular lesions, and 10 control non‐infected dogs. Complete macroscopic ophthalmic examination of eyelids and globes was performed. Both total and anti‐Leishmania infantum IgG antibodies were studied in sera and aqueous humour (AH) of all dogs by ELISA technique. A Goldmann–Witmer coefficient (C value) was calculated. Results The main ophthalmological findings were keratoconjunctivitis (71.7%; 38/53), hyperplasia of conjunctival lymphoid follicles (54.7%; 29/53), blepharitis (50.9%; 27/53) and uveitis (20.7%; 11/53). Ocular production of anti‐Leishmania infantum IgG was detected in 73.6% (39/53) of infected dogs. There was no correlation between the antibody levels in AH and sera of the same dog. The mean anti‐Leishmania infantum IgG in AH was higher in uveitis, followed by lesions affecting only the adnexa (p < 0.0001). The highest mean C values were observed for uveitis, conjunctivitis and keratitis. Conclusions Our findings suggest that production of anti‐Leishmania IgG in dogs infected with Leishmania infantum with ocular manifestations begin in situ and follows by a transfer of antibodies from the bloodstream to the AH.
