Objective: To develop coenzyme Q10 (co-Q10) nanostructured lipid carriers (NLCs) using stearic acid (SA) and various liquid lipids with different lipophilicity as well as highlights the use of in silico studies for predicting and elucidating the interaction of drug-lipid used as carries in NLCs, at the molecular level. Methods: The co-Q10 NLCs were prepared using SA as solid lipid and oleic acid (OA), isopropyl myristate (IPM), as well as isopropyl palmitate (IPP) as liquid lipids by the high shear homogenization method. Firstly, the formulas were optimized by the appropriate required HLB (rHLB). The optimized NLCs were characterized in the particle size, distribution of particle size, zeta potential, crystallinity behavior, Fourier transform infrared (FT-IR) spectra, morphology, entrapment efficiency (EE), drug loading (DL), and pH value. The interaction of drug-lipids in silico was studied using the AutoDock Vina program. Results: The co-Q10 NLCs using SA and the various liquid lipid possessed the mean particle size, polydispersity index (PDI), zeta potential, EE, DL, and pH values were 180 to 350 nm,<0.5,<-30 mV, 83 to 88%, 10 to 11%, and 5.0 to 5.6, respectively. The EE and DL of co-Q10 NLCs increased with decreasing in binding energy (∆G) in silico. Conclusion: The co-Q10 NLCs using SA as solid lipid and OA, IPM, as well as IPP as liquid lipids were developed successfully. Furthermore, in silico study by molecular docking is a potential approach in predicting and elucidating the interaction of drug-lipid in the development of NLCs formulation.

Objective: In the current study, the Quality by Design method was utilized for the formulation of solid lipid nanoparticles of Methotrexate (MTX SLNs). Methods: MTX SLNs formulated by melt emulsification method were studied for the effect of independent variables viz. concentration of lipid and surfactants on quality attributes viz. particle size, polydispersity index, and entrapment efficiency of SLNs using 32 factorial design. Results: The optimal formulation was spherical, had a particle size of 147.6±4.1 nm (z-average), a polydispersity index of 0.296±0.058, a zeta potential of −19±0.98 mV, encapsulation efficiency of 98.7±1.55%, and a cumulative drug release of 95.59±0.918% in 5 h. Conclusion: The in vitro and in vivo studies revealed that SLNs provide a promising oral delivery system to improve the bioavailability of MTX.

Background:The aim of this study was to enhance the solubility of Natamycin, an antifungal agent used in fungal keratitis, through solid dispersion incorporated ophthalmic films with the purpose to avoid frequent dosing, enhance the period of retention, and sustaining release for better patient compliance. Materials and Methods: Firstly, solid dispersions were prepared by the solvent evaporation method and then, the best solid dispersion formulation was incorporated into films which were prepared by the solvent casting method using retarding polymers Ethyl cellulose, PVA and Chitosan. PEG-400 was used as a plasticizer. The films were evaluated for their thickness, surface pH, drug content, weight uniformity, folding endurance, tensile strength, antimicrobial activity, in-vitro and ex-vivo permeation studies, and in-vitro antifungal studies. Results and Discussion: The films exhibited good mechanical properties with promising results and were in an acceptable range. The optimized formulation NF6 shows more drug release in in-vitro studies as compared to ex-vivo. This is due to the varying pore size of the cornea and disturbance of other tissues. An ocular irritation test revealed that the prepared formulation was non-irritant. Conclusion: Thus, the formulated ophthalmic film seems to be a promising formulation for the safe and effective delivery of Natamycin through the ocular route in the treatment of fungal keratitis.
