Ocimum gratissimum Essential Oil and Its Isolated Compounds (Eugenol and Myrcene) Reduce Neuropathic Pain in Mice

Abstract: Ocimum gratissimum is used in popular medicine to treat painful diseases. The antihypernociceptive properties of O. gratissimum essential oil and two of its active components (eugenol and myrcene) were tested in a model of neuropathic pain induced by a chronic constriction injury of the sciatic nerve. In tests to determine chronic antinociception, adult male C57BL/6 J mice were treated orally with corn oil (control group), O. gratissimum essential oil at doses of 10, 20, or 40 mg/kg or eugenol or myrcene at do… Show more

“…Myrcene has been identified as an analgesic in past studies including those of Traditional Medical System (TMS) approaches. As a component of Occimum oils, it has demonstrated anti-neuropathic pain effects in mice, working in combination with eugenol (a TRPA1 ligand) [65]. Analgesic oils from other settings contain Myrcene [66][67][68]69].…”

“…Ocimum gratissimum L. is widely found in several geographical regions in South America and Africa [ 198 , 199 ] and still used as a medicinal plant with analgesic activity [ 198 ]. It contains several proanthocyanidins, which have been shown to exhibit significant antioxidant activity, and tannins, saponins, steroids, alkaloids, terpenoids, flavonoids, phenols, and cardiac glycosides [ 200 ].…”

“…In addition, the administration of eugenol (1, 5 and 10 mg/kg) markedly decreased IL-1β levels whereas no significant effect was caused by treatment with myrcene. Similar antihypernociceptive activity was obtained with O. gratissimum essential oil (20 and 40 mg/kg) in the neurophatic pain models, providing evidence for the biological action that supports its popular use [ 79 ].…”

“…Eugenol is also the major constituent of the essential oil of Ocimum gratissimum L. (Lamiaceae), a plant popularly used in the treatment of painful diseases. In a model of neuropathic pain induced by chronic constriction of the sciatic nerve, the oral exposure to eugenol and the monoterpene myrcene (5 or 10 mg/kg, for 14 days after surgery) produced antihypernociceptive effects on male C57BL/6J mice tested in mechanical (von Frey) and thermal (hot plate) tests [ 79 ]. In addition, the administration of eugenol (1, 5 and 10 mg/kg) markedly decreased IL-1β levels whereas no significant effect was caused by treatment with myrcene.…”
