ABSTRACT. Bisphenol A (BPA) is an industrial contaminant and is reported to be a risk factor associated with the development of tumors. In our previous studies, we have shown that BPA promoted the growth of SK-N-SH human neuroblastoma cells and increased their invasion and metastasis. In this study, we further investigated the effects of BPA and 17b-estradiol (E 2 ) on the stem cell-like cells from SK-N-SH cells. Detection of stem cell markers, proliferation assay, and clonogenic analysis showed that the side-population (SP) of SK-N-SH cells had properties similar to those of stem cells. BPA or E 2 exposure decreased the percentage of SP cells and the expression of stem cell-marker proteins. BPA and E 2 promoted the growth of non-SP cells to a greater extent than of SP cells; in addition, they significantly increased the 2987 BPA promotes the growth of cancer stem cell-like SP cells ©FUNPEC-RP www.funpecrp.com.br Genetics and Molecular Research 14 (2): 2986-2993 (2015 growth of SP cells. WHAT IS KNOWN ALREADY:The few epidemiologic studies performed suggest that bisphenols have potential endocrine disruptive properties, but they did not identify clear and direct patterns of endocrine disruption. STUDY DESIGN, SIZE, DURATION:Adult human testis explants in culture were exposed to BPA and the analogs bisphenol F (BPF), bisphenol S (BPS), bisphenol E (BPE), bisphenol B (BPB), and bisphenol A diglycidyl ether (BADGE) at 10 -9 to 10 -5 M for 24 h or 48h.PARTICIPANTS/MATERIALS, SETTING, METHODS: Human adult testes were obtained from prostate cancer patients who had no hormone therapy, or from multiorgan donors. After ex vivo exposure to the investigated bisphenols, the measured outcomes were related to histopathology (gross morphology and germ cell viability determined by anti-caspase 3 immunohistochemistry), and the levels of testosterone, insulin-like fator 3 and inhibin B were measured using immunoassays. The levels of mRNA encoding key enzymes of bisphenol biotransformation were investigated by quantitative PCR: UGT2B15 UDP (glucuronosyltransferase two family, polypeptide B15), GUSB (glucuronidase beta), SULT1A1 and 3 (sulfotransferase family 1 A member 1 and 3) and STS (steroid sulfatase). MAIN RESULTS AND THE ROLE OF CHANCE: A significant dose-dependent inhibition wasfound between testosterone levels measured in the culture medium and concentrations of BPA The active concentrations of BPA and BPA-A used in the study were higher than those found in human biological fluids. WIDER IMPLICATIONS OF THE FINDINGS:Under our experimental conditions, direct exposure to BPA or BPA-A can result in endocrine disturbance in the adult human testis.

In the last five years laser cutting has been identified as one of the solutions for converting and shaping the plastic films used in the Thin Film Transistor Liquid Crystal Display (TFT-LCD) industry due to its positive effects on cost management. However, there has been quite limited information available with regard to the hazards of the air pollutants released during the laser cutting process, and in particular in relation to the real operations that occur in a factory. This is the first study that aims to assess the Endocrine Disruptors (EDs) and Particulate Matter (PM) emitted during the laser cutting process. Phthalate Esters (PAEs) and Polycyclic Aromatic Hydrocarbons (PAHs) are the representative EDs in this research. The results indicate that the power of the laser and type of plastic significantly affect the composition of air pollutants. Di(2-ethylhexyl) phthalate (DEHP) of PAEs was identified in the Laser Generated Air Contaminants (LGACs) from both Polycarbonate (PC) and Polyethylene Terephthalate (PET) films. DEHP was detected in the gas-phase LGACs of PC and PET at low, medium and high power (80 W, 160 W, 240 W), but not detected in the particle-phase LGACs of PC and PET, except when the laser was operated at a low power setting. With the laser power increased, the concentrations of PM and PAHs also increased. This research found that during laser cutting PC released more DEHP than PET, but PET released more PAHs than PC.
