NRP1 regulates HMGB1 in vascular endothelial cells under high homocysteine condition

Abstract: Endothelial inflammation plays an important role in hyperhomocysteinemia (HHcy)-associated vascular diseases. High mobility group box 1 (HMGB1) is a pro-inflammatory danger molecule produced by endothelial cells. However, whether HMGB1 is involved in vascular endothelial inflammation of HHcy is poorly understood. Neuropilin-1 (NRP1) mediates inflammatory response and activates mitogen-activated protein kinases (MAPKs) pathway that has been reported to be involved in regulation of HMGB1. The aim of this study w… Show more

“…And the molecular mechanism of VEGF-Nrp1 interaction is insufficiently reported [40]. Ma et al found that overexpression of Nrp1 promoted the expression and secretion of high mobility group box 1 (HMGB1) and endothelial inflammation, and that the mitogen-activated protein kinase (MAPKs) pathway was involved in this process [41]. Consistent with the above studies, we also found that NRP1 was highly expressed in GBM patients.…”

“…The collected serum was placed in a refrigerator at -80˚C for testing. The level of homocysteine (Hcy) was detected by cyclic enzyme method ( 19 ). The level of miR-28-3p was detected by real-time fluorescence quantitative PCR (qRT-PCR).…”

“…NRP1 is also a co-receptor for the VEGF signaling pathway, indicating a crosstalk between these two pathways. NRP1 also regulates HMGB1, which induces caveolae formation in general ECs (Ma et al, 2019). Possibly PDGF signaling is able to manage transcytosis via this pathway, additionally activating a regular PI3K/AKT pathway for actin dynamic regulation.…”
