Origin of muscle satellite cells in the Xenopus embryo

SUMMARYWe have studied the origin of muscle satellite cells in embryos of Xenopus laevis. Fate mapping at the open neural plate stage was carried out using orthotopic grafts from transgenic embryos expressing GFP. This shows that most satellite cells originate from the dorsolateral plate rather than from the paraxial mesoderm. Specification studies were made by isolation of explants from the paraxial and dorsolateral regions of neurulae and these also indicated that the satellite cell progenitors arise from the dorsolateral plate. Muscle satellite cells express Pax7, but overexpression of Pax7 in blastomeres of whole embryos that populate the myogenic areas does not induce the formation of additional satellite cells. Moreover, a dominant-negative construct, Pax7EnR, does not reduce satellite cell formation. Neither Pax7 nor other myogenic transcription factor genes will induce satellite cell formation in animal caps treated with FGF. However, BMP RNA or protein will do so, both for FGF-treated animal caps and for paraxial neurula explants. Conversely, the induction of Noggin in dorsolateral explants from HGEM-Noggin transgenic neurulae will block formation of satellite cells, showing that BMP signaling is required in vivo for satellite cell formation. We conclude that satellite cell progenitors are initially specified in the dorsal part of the lateral plate mesoderm and later become incorporated into the myotomes. The initial specification occurs at the neurula stage and depends on the ventral-to-dorsal BMP gradient in the early embryo.

The tail of the Xenopus tadpole will regenerate completely after transection. Much of the mass of the regenerate is composed of skeletal muscle, but there has been some uncertainty about the source of the new myofibres. Here, we show that the growing tail contains many muscle satellite cells. They are active in DNA replication, whereas the myonuclei are not. As in mammals, the satellite cells express pax7. We show that a domain-swapped construct, pax7EnR, can antagonize pax7 function. Transgenic tadpoles were prepared containing pax7EnR driven by a heat-inducible promoter. When induced, this reduces the proportion of satellite cells formed in the regenerate. A second amputation of the resulting tails yielded second regenerates containing notochord and spinal cord but little or no muscle. This shows that inhibition of pax7 action does not prevent differentiation of satellite cells to myofibres, but it does prevent their maintenance as a stem cell population.

Pax3 and Pax7 expression and regulation in the avian embryo

Satellite cells are essential for postnatal growth and repair of skeletal muscle. The paired-box transcription factors Pax3 and Pax7 are expressed in emerging muscle precursors. Recent studies have traced the origin of satellite cells to the embryonic dermomyotome, however, their developmental regulation throughout embryogenesis remains unclear. We show the overlying surface ectoderm and lateral plate are essential for Pax3 expression, and that the overlying surface ectoderm and neural tube are necessary for Pax7 expression within the dorsal somite. Furthermore we show that the notochord acts to down regulate the expression of both genes. Moreover, we identify diffusible factors within these tissues that act to maintain expression of Pax3 ( + ) and Pax7 (+) muscle precursors. We show that Wnt1, 3a, 4 and 6 proteins are able to up regulate and expand the expression of Pax3 and Pax7 within the dorsal somite. Finally, we show that Wnt6 can mimic the effect of the dorsal ectoderm to maintain Pax3 and Pax7 expression.
