Nitrogenous Constituents and Free Amino acid Content in the Seminal Plasma of Buffalo (Bos Bubalis) in Different Periods of the Year in Relation to Fertility

Abstract

Summary Semen of eight Murrah buffalo bulls was analysed for different nitrogenous constituents in different periods of the year and for the free amino acid content of seminal plasma. Total nitrogen, protein nitrogen, non‐protein nitrogen and amino acid nitrogen were found to be 649.10 ± 12.83, 532 ± 11.56, 116.26 ± 2 and 99.13 ± 4.4 mg. / 100 ml., of seminal plasma. All the nitrogen fractions vary significantly (P ≤ 0.01) between the bulls and between the periods. All the nitrogen fractions tended to be higher in colder periods as compared to the hot ones. Protein contributed 82 % of the total nitrogen. Conception rate was lowest during the hot period. NPN and amino acid nitrogen had a positive correlation with conception rate. Ten free amino acid‐aspartic acid, glutamic acid, serine, alanine, glycine, lysine, arginine, histidine and tyrosine (tryptophan) were identified by two dimensional TLC. Glutamic acid was the predominant amino acid (59.78 %) followed by glycine, serine, alanine and aspartic acid. Glutamic acid and glycine alone constituted 78.49 % of the total amino acids. Zusammenfassung Stickstoffhaltige Bestandteile und freie Aminosäuren im Samenplasma von Büffeln (Bos Bubalis) in Relation zur Fruchtbarkeit während verschiedener Jahreszeiten Bei 8 Murrah‐Büffeln wurde während verschiedener Jahreszeiten im Spermaplasma der Gehalt an stickstoffhaltigen Komponenten und freien Aminosäuren analysiert. Befunde (mg / 100 ml): Totalstickstoff 649,10 ± 12,83; Proteinstickstoff 532 ± 11,56; Nichtproteinstickstoff 116,26 ± 2; Aminosäurenstickstoff 99,13 ± 4,4. Alle N‐Fraktionen variierten signifikant sowohl von Tier zu Tier als auch von Jahreszeit zu Jahreszeit. Alle N‐Fraktionen zeigten in den kühlen Perioden steigende Tendenz im Vergleich zu den warmen Perioden. 82 % des N entfielen auf das Eiweiß. Während der warmen Periode war die Konzeptionsrate am kleinsten. Der NPN und Aminosäuren‐N zeigten positive Korrelationen zur Konzeptionsrate. Mittels der zweidimensionalen Dünnschichtchromatographie wurden 10 verschiedene Aminosäuren (Asparaginsäure, Glutaminsäure, Serin, Alanin, Glycin, Lysin, Arginin, Histidin, Tyrosin, Tryptophan) identifiziert. Die Glutaminsäure dominierte gegenüber den anderen Aminosäuren (59,78 %), gefolgt von Glycin, Serin, Alanin und Asparaginsäure. Glutaminsäure und Glycin machten 78,49 % aller Aminosäuren aus.
