This research was designed to evaluate the CNS depressant, anxiolytic, and analgesic action of aqueous and ethanol extract of Ganoderma applanatum , a valuable medicinal fungus used in multiple disorders belongs to Ganodermataceae family. Two extracts of G. applanatum were prepared using distilled water and ethanol as solvents and named AEGA and EEGA. Open field method, rotarod method, tail suspension method, and hole cross method were utilized for the CNS depressant action. In contrast, elevated plus-maze test and hole board method were utilized for the anxiolytic action. For determining the analgesic potential, acetic acid-induced writhing test, hot plate method, and tail immersion test were used. Besides, molecular docking has been implemented by using Discovery studio 2020, UCSF Chimera and PyRx autodock vina. At both doses (200 and 400 mg/kg) of AEGA and EEGA showed significant CNS depressant effect ( p < 0.05 to 0.001) against all four tests used for CNS depressant activity. Both doses of AEGA and EEGA exhibited important anxiolytic activity effect ( p < 0.05 to 0.001)against the EPM and hole board test. Both doses of AEGA and EEGA also exhibited a potential analgesic effect ( p < 0.05 to 0.001) against all three tests used for analgesic action. In addition, in the molecular docking the compounds obtained the scores of −5.2 to −12.8 kcal/mol. Ganoapplanin, sphaeropsidin D and cytosporone C showed the best binding affinity to the selected recptors. It can be concluded that AEGA and EEGA have potential CNS depressant, anxiolytic, and analgesic action, which can be used as a natural antidepressant, anxiolytic, and analgesic source.

Background: Diospyros malabarica is a well known flowering plant indigenous to Indian subcontinent which is used in folklore medicine for several purposes. Our study is designed to assess the neuropharmacological and antibacterial efficacy of the ethyl acetate extract of D. malabarica seeds. Methods: The behavioral anxiolytic activities of the extract were assessed by using open field (OFT), hole cross (HCT), elevated plus maze (EPZ), hole board (HBT), light dark test (LDT); and antidepressant activities through forced swimming (FST) and tail suspension test (TST). Antimicrobial potential was assessed through disc diffusion method. Results: In OFT and HCT, the extract treated groups significantly (*p < 0.05) decrease the movement of animals when compared to vehicle-treated group. Higher dose (400 mg/kg b.w.) of extract greatly increased the spending time in open arm of EPZ, which endorses anxiolytic-like behavior of extract. The observed effect may be due to binding of any phytoconstituent with GABA A receptor. HBT and LDT results support the exploratory behavior of mice. The extract significantly decreased the immobility time in FST (20.71% for 200 mg/kg, and 31.59% for 400 mg/kg extract) and TST, which indicates the occupancy of antidepressant-active constituents. Gram negative bacteria were susceptible to extract than Gram positive strains; however the antimicrobial effect is not significant, hence trivial to declare. Conclusion: Our study demonstrates the possession of significant anxiolytic and antidepressant effects of D. malabarica extract which could be helpful for drug development program. Before potential therapeutic use, finding of the exact phytoconstituents with their mechanisms, and clinical trial are recommended.

Background Schizophrenia (SCZ) is an incurable neuropsychiatric disorder generally described by impaired social behavior and altered recognition of reality. For the first time, this study explored serum levels of antioxidants (vitamin A, E, and C), malondialdehyde (MDA), macro‐minerals (calcium, potassium, and sodium), and trace elements (zinc, iron, and selenium) in Bangladeshi patients with SCZ and thereby, discovering any pathophysiological correlation. Methods This case‐controlled study evaluated 63 patients with SCZ as cases and 63 healthy individuals as controls. Vitamin A and E levels were defined by RP‐HPLC. MDA and vitamin C levels were measured by using UV spectrophotometry, and macro and trace elements by atomic absorption spectroscopy. Results This study found significantly ( P ≤ 0.05) elevated MDA levels and decreased levels of antioxidants—vitamin A, C, and E and significantly ( P ≤ 0.05) diminished levels of macro and trace elements in cases in contrast to the controls. Serum levels of zinc (Zn), selenium (Se), iron (Fe), potassium (K), calcium (Ca), and sodium (Na) were determined to be 0.33 ± 0.008, 0.0252 ± 0.00060, 0.24 ± 0.01, 64.18 ± 2.72, 36.88 ± 2.56, and 2657.5 ± 53.32 mg/L, respectively, in cases, whereas 0.79 ± 0.03, 0.0650 ± 0.00355,0.78 ± 0.03, 168.01 ± 2.85, 86.43 ± 2.55, and 3200.8 ± 29.96 mg/L, respectively, were determined in controls. Pearson's correlation analysis revealed a negative correlation between Zn and Na, Zn and K, Zn and Ca, Zn and Fe, Zn and Se, Fe and Na, and Fe and Se in patients. Conclusions The findings connect that the pathogenesis of SCZ may have a correlation with altered levels of antioxidants, MDA, macro‐minerals, and trace elements.
