mRNA analysis of several components of the plasminogen activator and matrix metalloproteinase systems in endometriosis using a real-time quantitative RT–PCR assay

Abstract: The increase in mRNA and protein levels of uPA and MMP-3 observed in endometrium of women with endometriosis may facilitate the attachment of endometrial tissue to the peritoneum and ovarian surface, as well as the invasion of the extracellular matrix. This process would lead to the formation of early endometriotic lesions. Once the ovarian endometriotic cyst is developed, PAI-1 and TIMP-1 would increase which could explain the frequent clinical finding of an endometrioma without invasion of the adjacent ovari… Show more

“…The establishment of ectopic sites of endometrial growth is an invasive event linked, among other things, to altered expression of MMPs. Gilabert-Estellés et al [17] showed that the mRNA expression of MMP-3 was significantly higher in the endometrium of women with endometriosis than in controls, confirming previous reports [27,28,29,30]. Our data indicate that the mRNA level of MMP-3 was significantly higher in peripheral blood from endometriotic women than from control samples.…”

“…MMPs act in dynamic morphogenetic processes degrading and remodeling extracellular matrix, stimulating cell proliferation and apoptosis and inducing cell migration. Several previous studies have suggested that eutopic and ectopic endometrium from women with endometriosis shows altered expression levels of the MMP system, including MMP-3 and MMP-9, indicating that these two enzymes play a role in the pathogenesis of this disease [17,27,28,29,30,31,32]. Scant attention has been paid to the identification of serum levels of MMP-3 and MMP-9 in women with or without endometriosis.…”

“…The amplicon lengths of the QPCR assays for uPA and PAI-1 were minimized as far as possible to potentially enable quantification of partially fragmented mRNA extracted from formalin-fixed, paraffinembedded tissue (38). Recently, several QPCR assays for determination of uPA and/or PAI-1 mRNA expression have been introduced with rather large amplicon sizes (13)(14)(15)17,18,39,40). In contrast, our newly developed QPCR assays use amplicon lengths ranging from 103 to 132 base pairs, the shortest uPA/PAI-1 QPCR-derived cDNA fragments described so far.…”
